Determination of toxic elements and their species in clinical materials

Abstract:

V této diplomové práci se věnujeme analýze forem toxických prvků v klinických materiálech s důrazem na stanovení methylrtuti v mateřském mléce. V teoretické části práce se zabýváme vlastnostmi, biogeochemickým cyklem rtuti, kadmia, olova a arsenu, toxicitou jejich specií, vzorkováním a uchováváním klinických vzorků, možnostmi stanovení celkového obsahu těchto toxických prvků a stanovením methylruti v klinických vzorcích (krev, mateřské mléko, svaly, vlasy). V experimentální části práce bylo pro stanovení koncentrace methylrtuti ve vzorcích mateřského mléka využito spojení metod vysokoúčinné kapalinové chromatografie a hmotnostní spektrometrie s indukčně vázaným plazmatem. Pomocí standardních roztoků byly zjištěny základní analytické parametry stanovení, jako jsou separace specií, linearita, limit detekce, a další. Vhodnost metody pro stanovení methylrtuti byla dále ověřena na vzorcích kravského mléka a poté byla metoda aplikována na reálné vzorky mateřského mléka a byly zjištěny další parametry stanovení: opakovatelnost a výtěžnost. Poté byla metoda aplikována na vzorky mateřského mléka norské kohorty projektu DENAMIC. Také byla vyvíjena metoda pro zakoncentrování a přečištění vzorku.

Abstract:

In this thesis we deal with the determination of toxic elements and their species in clinical materials with an emphasis on determination of methylmercury in breast milk. In theoretical part we deal with properties, biogeochemical cycle of mercury, cadmium, lead and arsenic and toxicity of their species. Furthermore, we presented sampling and storage of clinical samples and possibilities of measuring total mercury, cadmium, lead, arsenic and methylmercury determination in clinical samples (blood, breast milk, urine, fingernails, muscles, scalp hair). High-performance liquid chromatography coupled to mass spectrometry with inductively coupled plasma was used for determination of methylmercury within the samples. Basic analytical parameters, such as separation of species, linearity and limit of detection were determined using standard solution of methylmercury. Suitability of this method for the determination of methylmercury was further validated on cow milk and then on breast milk samples. Subsequently repeatability and recovery of this method were determined. This method was also applicated on real samples of breast milk from norway's cohort (DENAMIC project). The method for preconcentration and purification of the sample was also developed.
