Electrochemical study of nucleic acids and their components by elimination voltmetry

Abstract:

V předložené dizertační práci byly pomocí adsorptivní rozpouštěcí technikou ve spojení s eliminační voltametrií (Elimination voltammetry with linear scan – EVLS) studovány krátké řetězce deoxyribonukleových kyselin – nonamery, dva homo-ODN dA9, dC9 (5´-AAA AAA AAA- 3´,5´-CCC CCC CCC- 3´) a tři hetero ODN H3, H4, H9 (5´- CCC AAA CCC - 3´,5´- CAC CAC CAC - 3´, 5´- ACC CAC CCA - 3´). Po aplikaci EVLS funkce E4 (eliminace nabíjecího a kinetického proudu, zachování proudu difúzního) došlo u ODN ke vzniku typických, podle teorie potvrzených, signálů ve tvaru pík-protipík, které zaručovaly několikanásobné zvýšení jejich intenzity. U studovaných hetero-ODN bylo za vhodných podmínek možné pomocí EVLS rozdělit CA pík na dva redukční signály odpovídající bázím A a C. Dále bylo zjištěno, že separace CA píku je ovlivněna celou řadou parametrů. Nejvýznamnějším z nich bylo pH a sekvence ODN, méně významná byla teplota a doba akumulace. V závislosti na nich byla eliminační voltametrie schopná odhalit dynamiku procesů probíhajících na povrchu elektrody. Povrchové vlastnosti ODN jsou úzce spjaty s chováním ODN v roztoku. Proto byla elektrochemická studie doplněna experimenty cirkulárního dichroismu a gelové elektroforézy, které potvrdily, že eliminační signály A a C jsou ovlivněny nejen primární, ale i sekundární strukturou homo- i hetero-ODN. Dílčí cíl práce, týkající se aplikace metody konfidenční elipsy jako součást statistické analýzy, byl realizován nejen v elektrochemii, ale i ve výše uvedených doplňkových metodách. Teoreticky je dvojnásobná eliminace zatížena velkou chybou, a proto pro nás bylo překvapující, že statistické vyhodnocení experimentálních dat ukázalo relativní chybu analýzy maximálně do 5 %. Námi získané výsledky naznačují velký potenciál EVLS ve spojení s adsorptivní rozpouštěcí technikou pro vývoj a výzkum ODN senzorů nejen s využitím rtutí modifikovaných elektrod, ale i pevných elektrod.

Abstract:

In the present PhD thesis, short-chain deoxyribonucleic acids (nonamers), two homo-ODN dA9, dC9 (5'-AAA AAA AAA-3', 5'-CCC CCC CCC-3'), and three hetero-ODN H3, H4, H9 (5'-CCC AAA CCC-3', 5'-CAC CAC CAC-3', 5'-ACC CAC CCA- 3', respectively) were studied by adsorptive stripping technique in connection with elimination voltammetry (Elimination voltammetry with linear scan - EVLS). When applying EVLS function E4 (eliminating kinetic and charging currents, and conserving diffusion current), typical, peak-counterpeak signals confirmed according to the theory, were observed. The heights of these signals are multiple times higher than the voltammetric peaks measured. In the hetero-oligonucleotides studied, the CA reduction peak can, under optimized conditions, be separated into two EVLS signals corresponding to the single nucleic bases (A and C). It was found that the separation of the CA peak is influenced by a number of parameters. The most important of these were pH and the ODN sequence, minor ones were the temperature and time of accumulation. Depending on these experimental conditions, elimination voltammetry was capable of detecting the dynamics of the processes on the electrode surface. Surface properties of ODNs are closely associated with the behavior of ODNs in solution. Therefore, electrochemical studies were complemented by experiments with circular dichroism and gel electrophoresis, which confirmed that the elimination signals A and C are influenced not only by the primary but also by the secondary structure of homo- and hetero-ODN. In addition, the particular aim of the thesis, the application of the confidence ellipse method as part of statistical analysis, was realized not only in electrochemistry, but also in the above-mentioned additional methods. Theoretically, the double elimination is burdened with high deviation. Thus, it was surprising for us that the statistical evaluation of the experimental data analysis showed a relative deviation below 5 %. Our results showed a great potential of EVLS in connection with adsorptive stripping voltammetry for the research of ODN by sensors not only using modified mercury electrodes, but also solid electrodes.
