Abstract:

Staphylococcus aureus je oportunní patogen, který se u části populace vyskytuje jako běžný kolonizátor nosní sliznice. Problém představuje jeho zvyšující se rezistence na používaná antibiotika, zejména meticilin. Tyto meticilin-rezistentní kmeny, jež jsou zvláště aktuálním problémem v nemocničním prostředí, mohou způsobovat až fatálně probíhající infekce. Proto je zájem o nalezení alternativního způsobu léčby těchto infekcí a jako jedna z perspektivních možností se jeví fágová terapie. Terapeutické využití bakteriofágů přináší oproti antibiotikům řadu výhod, jako například malou pravděpodobnost vzniku rezistence. Výzkum bakteriofágů začal již počátkem minulého století, záhy ale utichl díky objevu antibiotik, která se začala masově rozmáhat. V poslední době se zájem o tuto problematiku opět obnovuje z důvodu vzrůstající rezistence na antibiotika. Tato diplomová práce se zabývá nalezením a charakterizací mutant polyvalentních stafylokokových fágů, které by byly schopny lyzovat širší spektrum především meticilin-rezistentních kmenů Staphylococcus aureus, a tím by byly perspektivní v terapeutickém využití. Bylo izolováno pět nových mutant, u kterých bylo ověřeno jejich širší lytické spektrum a provedena molekulárně-biologická charakterizace.

Abstract:

Staphylococcus aureus is an oportunistic pathogen present in a part of the human population as a common nose mucosa colonizer. Problematic is its raising resistance to some of the antibiotics currently in use and especially methicillin. These methicillin-resistant strains are a particular problem in hospital environments where they can cause even fatal infections. Therefore there is a big interest in finding alternative therapies for these infections and one of the perspective possibilities seems to be phage therapy. The therapeutic utilization of bacteriophages has several advantages over usage of antibiotics, for example a smaller chance of resistance being formed. The research of bacteriophages began in the early 20th century but it soon came to a halt with the emergence of antibiotics which were undergoing a mass expansion at the time. Interest in these issues has been restored lately due to the raising resistance to antibiotics. This diploma thesis is concerned with finding and characterizing staphylococcal host-range mutants which would be able to lyse a wider spectrum of prevalently methicillin-resistant strains of Staphylococcus aureus and thus be perspective in therapeutic utilization. There were isolated five new host-range mutants in which was confirmed broader lytic spectrum and they were characterized by molecular techniques.
