Preparation of multiplex CRISPR-KO vectors for mutagenesis of alfalfa (Medicago sativa L.) MAP kinase

Abstract:

Lucerna siata (Medicago sativa L.) patrí medzi najvýznamnejšie plodiny sveta využívané v mnohých odvetviach. Dôležitosť výskumu tohto poľnohospodársky nenahraditeľného druhu, viedla k štúdiu mitogénom aktivovaných proteín kináz (MAPK), ktoré v bunke regulujú mnohé transkripčné faktory, cytoskeletálne a ďalšie dôležité proteíny. MAPK zohrávajú kľúčovú úlohu počas rastu a vývinu rastlín ako aj pri odpovediach na zmeny v okolitom prostredí. Medzi tieto enzýmy patrí MMK2 (Medicago MAPK2), ktorá okrem účasti na signalizácii počas reakcií na stresy biotického a abiotického pôvodu, ovplyvňuje aj organizáciu cytoskeletu a biosyntézu bunkovej steny. Z doteraz publikovaných imunolokalizačných štúdií vyplynula hypotéza o možnom zapojení MMK2 počas interakcií lucerny s prospešnými baktériami. Pokiaľ ide o riešenie moderných problémov, ako je rastúci dopyt po potravinách a plodinách v meniacom sa prostredí, funkčné štúdie génu MMK2 môžu poskytnúť priestor pre ďalší výskum. Pre tento účel je nevyhnutné pripraviť transgénne rastliny s vyradením (knock-out, KO) príslušného génu. Predkladaná práca je preto zameraná na prípravu destinačných multiplexných CRISPR-KO vektorov pre mutagenézu, prakticky, akéhokoľvek génu. Pripravené boli rôzne typy destinačných vektorov, líšiacich sa systémom selekcie a expresiou nukleázy Cas9. Berúc do úvahy zložitejší spôsob získavania ďalších generácií lucerny, okrem konštitutívnej expresie Cas9, bol pripravený aj variant s inducibilným systémom. Do výsledných destinačných vektorov možno naklonovať 1 až 12 rôznych vodiacich RNA (gRNA) pre nasmerovanie mutagenézy. V rámci predkladanej bakalárskej práce bolo navrhnutých viacero gRNA pre mutagenézu génu MMK2, vrátane dizajnu primerov pre ich budúce klonovanie do príslušných akceptorových vektorov.

Abstract:

Abstract Alfalfa (Medicago sativa L.) is one of the most important crops globally used in many fields of industry. The importance of research on this agriculturally irreplaceable species has led to the study of mitogen-activated protein kinases (MAPKs), which regulate many transcription factors, cytoskeletal and other essential proteins. MAPKs play a key role during plant growth and development and in responding to changes in the environment. These enzymes include MMK2 (Medicago MAPK2) which, besides participating in signalling during stress responses of biotic and abiotic origin, affects the cytoskeletal organisation and cell wall biosynthesis. The published immunolocalisation studies have emerged a hypothesis about the possible involvement of MMK2 during alfalfa interactions with beneficial bacteria. Concerning solving modern problems, such as the increased demand for food and crops in a changing environment, functional studies of the MMK2 gene can provide scope for further researchs. For this purpose, it is necessary to prepare transgenic plants with knock-out (KO) of this gene. Therefore, the present thesis focuses on the preparation of destination multiplex CRISPR-KO vectors for mutagenesis of practically any gene. Different types of destination vectors have been prepared that differ in the selection system and expression of the Cas9 nuclease. Considering difficulties in producing the next alfalfa generation, in addition to the constitutive expression of Cas9, a variant with an inducible system was prepared. One to twelve different guide RNAs (gRNAs) can be cloned into the final destination vectors to direct mutagenesis. Within this bachelor thesis, several gRNAs for MMK2 gene mutagenesis have been proposed, including the design of primers for their future cloning into appropriate acceptor vectors.
