Antimicrobial resistance and its determinants in bacteria isolated from ornamental fish

Abstract:

Šíření antimikrobiální rezistence v důsledku neuváženého používání antimikrobiálních látek se stalo v posledních letech vážným celosvětovým problémem nejen v humánní, ale i ve veterinární medicíně. S rezistencí patogenních bakterií, následným selháním terapie a vznikem vážných ekonomických ztrát se potýkají i akvakultury. Zvláštní skupinou takto chovaných organizmů jsou akvarijní ryby. Studií zabývajících se rezistencí bakterií izolovaných z akvarijních ryb bylo do současné doby provedeno velmi málo. V rámci této diplomové práce bylo identifikováno celkem 118 izolátů z akvarijních ryb importovaných nejčastěji z jihovýchodní Asie. Jejich citlivost k 12 antimikrobiálním látkám byla stanovena difuzní diskovou metodou a u bakterií rodu Aeromonas i agarovou diluční metodou. Bylo zjištěno vysoké procento izolátů rezistentních zejména k ampicilinu, oxytetracyklinu a kyselině oxolinové. Jednotlivé izoláty vykazovaly rezistenci současně k několika testovaným antimikrobiálním látkám (u rodu Aeromonas nejčastěji k 9, u rodu Pseudomonas dokonce k 11 a u kategorie Ostatní ke 4). U vybraných aeromonád, indikátorového rodu pro monitoring antimikrobiální rezistence ve vodním prostředí, byly hledány determinanty rezistence k sulfonamidům (sul1, sul2, sul3 geny) metodou PCR. Nejvíce testovaných izolátů neslo gen sul1 (46 z 57), gen sul2 byl přítomen u 10 izolátů, gen sul3 nebyl detekován vůbec. Vzhledem k výskytu genu sul1, a tudíž i riziku přítomnosti integronů, které významným způsobem přispívají k horizontálnímu přenosu genů kódujících antimikrobiální rezistenci u gramnegativních bakterií, byla u izolátů pozitivních na uvedený gen provedena PCR reakce na průkaz integronů (int1 a int2 gen) a genových kazet. Byly detekovány integrony třídy 1 a genové kazety aadA1, aadA2, dhfr1 a dhfr12.

Abstract:

In recent years, the emergence of antimicrobial resistence as a consequence of the inappropriate application of antimicrobial compounds has become a serious problem worldwide not only in human, but also in veterinary medicine. The aquaculture industry has also been affected by the incidence of resistant pathogenic bacteria, followed by treatment failure and serious economic losses. Ornamental fish are a special group of aquaculture animals. There is a lack of publications describing the resistance of bacteria isolated from ornamental fish. In this master thesis, 118 isolates from ornamental fish imported mostly from southeast Asia have been identified. Their susceptibility to 12 antimicrobial compounds has been determined by difussion disk method, and bacteria from Aeromonas sp. were also tested by agar dilution method. A high percentage of ampicilin-, oxytetracycline- and oxolinic acid-resistant bakteria were found. Single isolates have demonstrated simultaneous resistance to several tested compounds (Aeromonas sp. most commonly to 9, Pseudomonas sp. even to 11 and the group „Others“ to 4). In the selected aeromonads - indicator species for monitoring of antimicrobial resistance in aquatic environment - resistance determinants to sulfonamides (genes sul1, sul2 and sul3) have been found by PCR method. Most of tested isolates have carried the sul1 gene (46 from 57), the sul2 gene has been found in 10 isolates, the sul3gene has not been detected. With regard to the incidence of the sul1 gene, and the risk of occurrence of the integrons, which are together with another elements recognised for the spread of genes coding antimicrobial resistance in gramnegative bacteria, isolates which were positive for the presence of the mentioned gene have been tested by PCR reaction for evidence of integrons (genes int1 and int2) and gene cassettes. Class 1 integrons and gene cassettes aadA1, aadA2, dhfr1 and dhfr12 have been detected.
