Abstract:

Vliv metabolického stavu buňky na vnímavost k apoptóze indukované oxidačním stresem: studie na buněčné linii K-562 Cíl studie: Zjistit zda metabolický stav buňky ovlivňuje její náchylnost k apoptóze vyvolané oxidačním stresem a zda je takto indukovaná apoptóza spouštěna prostřednictvím mitochondrií. Typ studie: Studie na buněčné linii in vitro. Materiál a metody: Buňky K-562 byly kultivovány v médiu RPMI 1640 a udržovány v exponenciální fázi růstu. Pro vlastní experiment byly buňky zcentrifugovány a resuspendovány v experimentálním médiu připraveném předem na bázi Krebsova média. Experimentální médium bylo obohaceno 5, 11, nebo 30 mM glukózou, nebo 5 mM glutaminem nebo 5 mM fruktózou coby energetickým substrátem. Po osmihodinové inkubaci v těchto médiích byly buňky vystaveny 20 min působení tert-butylhydroperoxidu (tBH) v koncentraci 250 nebo 500 µM; kontrolní skupina nebyla tBH vystavena. Poté byly buňky zcentrifugovány, propláchnuty v čerstvém mediu RPMI a inkubovány v mediu RPMI po 12 hodin. Pro zjištění vlivu dalších agens na toxicitu tBH byl užit stejný protokol, pouze koncentrace glukózy byla vždy 11 mM a koncentrace tBH 500 nebo 1000 µM. Byl testován vliv antioxidantu N-acetyl cysteinu v koncentracích 1, 2 a 4 mM, dále mitochondriálních toxinů rotenonu (12,5 μM) a cyklosporinu A (2 μM) a inhibitoru všech kaspáz Z-VAD-FMK (50 μM). K detekci apoptotických buněk bylo použito průtokové cytometrie v dvojitém barvení FITC-Annexinem V a propidium jodiem. Cytometrie byla provedena na přístroji Becton Dickinson FACScan, k analýze dat byl použit software „CellQuest“ firmy Becton Dickinson. Účinná koncentrace rotenonu byla určena spektrofotometrickým stanovením změn poměru laktátu a pyruvátu, zatímco k určení účinné koncentrace cyklosporinu A bylo použito stanovení mitochondriální retence kalcia fluorescenční spektrofotometrií. Vliv vysokých koncentrací glukózy na oxidativní poškození DNA byl zkoumán technikou DNA komet. Výsledky: tBH měl jasný toxický a na dávce závislý efekt, který byl oslabován N-acetylcysteinem. Vyšší koncentrace glukózy signifikantně (p=0,007) zvyšovala toxicitu tBH. Záměna glukózy jako energetického substrátu za 5 mM glutamin nevedla k statisticky významným změnám v počtu přežívajících buněk, 5 mM fruktóza však měla aditivní toxický efekt při vysoké koncentraci tBH. Rotenon ani cyklosporin A neměly žádný vliv na toxicitu tBH, ta však byla téměř úplně potlačena kaspázovým inhibitorem Z-VAD-FMK. Buňky inkubované v 30 mM glukóze vykazovaly statisticky signifikantně vyšší poškození DNA, než buňky inkubované v 5 mM glukóze. Závěr: Využívaný energetický substrát, a tím navozený metabolický stav buňky, výrazně ovlivňuje vnímavost buněk vůči apoptóze vyvolané oxidačním stresem. V tomto modelu apoptóza vyvolaná oxidačním stresem nevyžaduje spuštění mitochondriální cesty a je zřejmě zprostředkována přímou aktivací kaspáz. Vysoká koncentrace glukózy v mediu zvyšuje oxidativní poškození DNA.

Abstract:

The metabolic state of the cell and the susceptibilty to oxidative-stress-induced apoptosis: a K-562 cell line study. Objective: To examine whether the susceptibility of the cell to oxidative-stress-induced apoptosis may be affected by metabolic state of the cell, and to assess the role of mitochondria in this interaction. Study design: In vitro cell line study. Material and Methods: The K-562 cells were cultured in RPMI-1640 medium and maintained in the exponential growth phase. For the experiment, the cells were harvested by centrifugation and resuspended in experimental medium prepared individually. The experimental medium did not contain any organic substrate and was supplied with either 5 or 11 or 30 mM glucose, or 5 mM fructose, or 5 mM glutamine. An 8-hrs incubation in these media was followed by 20 min exposure to 250 or 500 μM tert-butylhydroperoxide (tBH) with a control group not exposed to tBH. The cells were thereafter washed with fresh RPMI, and incubated overnight for 12hrs. The same experiment layout was used to determine the effect of other agents on tBH toxicity, except that the experimental medium was always supplied with 11 mM glucose and the concentrations of tBH were 500 and 1000 uM. An antioxidant N-acetylcysteine was used in order to determine, whether tBH toxicity is bound to oxidative stress; then mitochondrial toxines rotenone (12,5μM) and cyclosporine A (2μM) and pan-caspase inhibitor Z-VAD-FMK (50μM) were tested. Flow cytometry in double staining with FITC-Annexin and Propidium iodide was used to detect apoptotic cells. A Becton Dickinson “FACScan“ analyzer with “CellQuest“ BD acquisition/analysis software was used in this study. The effective concentration of rotenone was estabilished by spectrophotometric measurement of changes the lactate/pyruvate ratio while that of cyclosporine A was estabilished using fluorescence spectroscopy „calcic capture“ technique. „DNA comets“ assay was used to determine the effect of high glucose concentration on DNA oxidative damage. Results: We observed clear dose-dependent toxic effect of tBH while N-acetylcysteine, a potent antioxidant, prevented it. We proved (p=0.007) that higher concentrations of glucose add to the cytotoxicity of tBH. We found no significant change if 5mM glutamin was used compared to 5mM glucose, but in 5mM fructose the toxicity of high tBH concentrations was even more expressed. Rotenone and cyclosporine A had no effect on tBH toxicity, on the other hand, adding a pan-caspase inhibitor Z-VAD-FMK prevented the cells almost completely from tBH-induced cell death. We found significantly greater DNA damage in cells incubated with 30 mM glucose compared to 5 mM glucose. Conclusion: The substrate utilized as energy source markedly influences cell’s susceptibility to oxidative-stress-induced apoptosis. In this model, initiation of mitochondrial pathway is not essential for development of apoptosis, which is probably triggered by direct caspase activation. High glucose concentration contributes to DNA damage.
