Gel electrophoresis as a tool for the detection of 3´- DNA ends prolonged by terminal deoxynucleotidyl transferase.

Abstract:

Apoptóza je charakteristická morfologickými změnami, které jsou úzce spojené s kaskádou biochemických a fyziologických událostí vedoucích ke ztrátě životaschopnosti buňky. Apoptóza může být indukovaná signály generovanými přímo v buňce nebo vazbou na receptor na povrchu buněk. Existují tři základní dráhy, jejichž spuštění vede k aktivaci proteolytické kaskády a k degradaci buňky. Apoptóza také hraje klíčovou roli v patogenezi mnoha onemocnění. K patologické inhibici apoptózy dochází u nádorových onemocnění, naopak patologická indukce vede k rychlejšímu rozvoji některých onemocnění, např. AIDS a neurodegenerativních chorob. K detekci apoptózy existuje několik standardních technik, jako je světelná a elektronová mikroskopie, TUNEL test a detekce fosfatidylserinu. Cílem diplomové práce bylo využít gelovou elektroforézu k detekci 3´- konců DNA. K fragmentaci plazmidu pBluescript SK (-) byly využity restrikční endonukleázy a fragmenty s 3´- OH přesahujícím koncem byly prodlouženy terminální deoxynukleotidyl transferázou nukleotidy adeninu. Také byly využity magnetické částice oligo (dT)25, které obsahují oligo (dT) sekvenci vázanou na povrchu částic. K detekci volných konců byla použita i nylonová membrána. Nakonec byla apoptóza indukovaná peroxidem vodíku u kvasinek Saccharomyces cerevisiae a předchozí pokusy byly aplikovány na eukaryotní DNA.

Abstract:

Apoptosis is characterized by morphological changes, which are closely associated with a cascade of biochemical and physiological events leading to loss of cell viability. Apoptosis can be induced by signals generated in the cell or by binding to a receptor on the cell surface. There are three paths whose execution results in the activation of proteolytic cascades and degradation of the cell. Apoptosis also have a very important role in the pathogenesis of many diseases. The pathological inhibition of apoptosis occurs in cancer, while pathological induction leads to faster development of certain diseases, such as AIDS and neurodegenerative diseases. To detect apoptosis in several standard techniques, for example light and electron microscopy, TUNEL assay and detection of phosphatidylserine. The main point thesis was to use gel electrophoresis to detect 3' - ends of DNA. The fragmentation of the pBluescript SK(-) were used restriction endonucleases and the fragments with 3'- OH ends were prolonged terminal deoxynucleotidyl transferase adenine nucleotides. Also, the magnetic particles were used oligo (dT)25, which contain oligo (dT) sequence bound to the particle surface. To detect free ends was used as the nylon membrane. Finally, the apoptosis induced by hydrogen peroxide in the yeast Saccharomyces cerevisiae and previous attempts have been applied to eukaryotic DNA.
