Abstract:

Reakcí 3-brom-1 benzofuran-2(3H)-onu s thiomočovinou a jejími N substituovanými (methyl, isopropyl, terc-butyl, fenyl), N,N'-disubstituovanými (dimethyl, difenyl) a N,N,N'-trisubstituovanými (dimethyl-fenyl a dimethyl-4 methoxyfenyl) deriváty bylo připraveno 9 původních isothiouroniových solí 1a-i. Tyto relativně nestálé soli byly charakterizovány 1H a 13C NMR spektry, bodem tání a elementární analýzou. Přesmykem 7 solí v prostředí vodných aminových pufrů vznikají odpovídající substituované 2-imino-1,3 thiazolidin-4-ony 2a-g, jejichž struktura byla rovněž ověřena spektrálními metodami. Z 1H NMR spekter je patrné, že 2-alkylimino i 2 fenylimino deriváty 2b-e se v roztoku hexadeuteriodimethylsulfoxidu nachází ve směsi dvou tautomerů lišících se polohou dvojné vazby C=N (endo- resp. exocyklické), kdy tautomer s exocyklickou dvojnou vazbou dále existuje ve formě dvou geometrických izomerů s rozdílnou konfigurací. Při zahřátí na 60 °C lze pozorovat vzájemnou přeměnu těchto isomerů. Dále byla studována kinetika a mechanismus přesmyku N-fenyl-S-(2-oxo-2,3 dihydro-1-benzofuran-3 yl)isothiouronium-bromidu (1e) na 5-(2-hydroxyfenyl)-2 (fenylimino)-1,3-thiazolidin-4-on (2e) ve vodných roztocích tris(hydroxymethyl)aminomethanového (TRIS), N-methylmorfolinových (NMF) a N methyldiethanolaminového (MDEA) pufru v rozmezí pH 7,7 - 9 při 25 °C a I = 1 mol l-1 za podmínek pseudoprvního řádu. Ze změřených závislostí bylo zjištěno, že přesmyk je obecně kysele katalyzován, avšak prakticky nezávisí na pH. Rychlost určujícím krokem přesmyku je rozpad neutrálního tetraedrálního intermediátu (T0), který je katalyzován kyselou složkou pufru [BH+].

Abstract:

Nine original substituted isothiuronium salts 1a-i were prepared by the reaction of 3-bromo-1 benzofurane-2(3H)-one with thiourea and its N-substituted (methyl, isopropyl, tert-butyl, phenyl), N,N'-disubstituted (dimethyl, diphenyl) and N,N,N'-trisubstituted (dimethyl-phenyl and dimethyl-4-methoxyphenyl) derivatives. These relatively unstable salts were characterized by 1H and 13C NMR spectra, melting point and elemental analysis. The rearrangement of seven above-mentioned isothiuronium salts in aqueous amine buffers possesses substituted 2-imino-1,3 thiazolidin-4-ones 2a-g whose structure was also determined by spectral methods. From the 1H NMR spectra, it is evident that 2 alkylimino as well as 2-phenylimino derivates 2b-e occur as a mixture of two tautomers differing in the configuration of the double bond C=N (endo- or exocyclic) in the solution of a hexadeuteriodimethylsulphoxide. The tautomer with exocyclic double bond exists in the form of two geometric isomers with different absolute configuration. The mutual interchange of these isomers is observable under heating (60 °C). Next, the kinetics and the mechanism of the rearrangement of N-phenyl-S (2-oxo-2,3-dihydro-1-benzofurane-3 yl) isothiuronium bromide to 5 (2 hydroxyphenyl)-2-(phenylimino)-1,3-thiazolidin-4-one (2e) were studied in aqueous tris(hydroxymethyl)aminomethane (TRIS), N-methylmorpholine (NMF) and N methyldiethanolamine (MDEA) buffers within a pH range from 7.7 to 9 at 25 °C and ionic strength I = 1 mol l-1 under pseudo-first order conditions. From measured dependencies it was found that the rearrangement is subject to general acid catalysis but it is virtually independent on pH of the solution. From these observations it was concluded that the rate-limiting step involves general acid-catalyzed decomposition of the neutral tetrahedral intermediate (T0) under action of the acid buffer component [BH+].
