Synthesis and study of the platinum complexes with 4-azaindole derivatives.

Abstract:

Byla připravena série platnatých koordinačních sloučenin obecného složení cis-[Pt(nAza)2I2] (komplexy 1 a 2) a cis-[Pt(NH3)(nAza)X2] (komplexy 3-5), kde nAza = 4-azaindol nebo N1-isopropyl-4-azaindol, a X= Cl nebo I. Připravené komplexy byly analyzovány elementární analýzou, nukleární magnetickou rezonancí (1H, 13C a 15N NMR), hmotnostní spektrometrií s ionizací elektrosprejem, infračervenou spektroskopií a simultánní TG/DSC termickou analýzou. nAza ligand se na centrální atom koordinuje přes N4 atom pyridinového cyklu. U připravených cis-dijodoplatnatých komplexů 1-4 byla metodami 1H NMR a ESI+ MS studována hydrolýza a interakce s modelovými biomolekulami (redukovaný glutathion, guanosin monofosfát), přičemž výsledky poukazují na odlišné chování obou typů studovaných komplexů (symetrické komplexy 1 a 2 vs. asymetrické komplexy 3 a 4).Z výsledků testování in vitro cytotoxicity proti lidským nádorovým buňkám karcinomu vaječníku (A2870) plyne, že symetrické komplexy 1 a 2 jsou účinnější než jejich asymetrické analogy (3 a 4), a dále že komplexy s N1-isopropyl-4-azaindolem (2 a 4) vykazují vyšší aktivitu než komplexy s 4-azaindolem (1 a 3). Nejnižší hodnotu, a to IC50 = 3,8+-0,8 mikroM, vykazoval komplex 2 (IC50 = 26,7+-1,8 mikroM pro cisplatinu).

Abstract:

A series of Pt(II) coordination compounds of the general composition cis-[Pt(nAza)2I2] (1 and 2) and cis-[Pt(NH3)(nAza)X2] (3-5), where nAza = 4-azaindole or N1-isopropyl-4-azaindole, and X= Cl, or I. The complexes were characterised by elemental (C, H, and N) analysis, nuclear magnetic resonance (NMR), mass spectrometry with electrospray ionization, infrared spectroscopy, and simultaneous TG/DSC thermal analysis. An nAza ligands coordinate to the central atom via the N4 atom of the pyridine cycle. The results of the studies (1H NMR and ESI+ MS) of hydrolysis and the interaction with model biomolecules (reduced glutathione and guanosine monophosphate), performed for the prepared cis-diiodoplatinum(II) complexes 1-4, showed different behaviour of both types of complexes (i.e. symmetric complexes 1 and 2 vs. asymmetric complexes 3 and 4). The results of the testing of in vitro cytotoxicity against human cancer ovarian cells (A2780) showed that symmetric complexes 1 and 2 are more effective than their asymmetric analogues. Moreover, the complexes with N1-isopropyl-4-azaindole (2 and 4) showed higher activity than complexes with 4-azaindole (1 and 3). The complex 2 was found to be the most effective with IC50 = 3,8+-0,8 microM (IC50 = 26,7+-1,8 microM for cisplatin)
