Analysis of the SMC5/6 complex subunit interactions

Abstract:

Complex SMC5/6 participates in post-replication DNA repair, in stabilization of stalled replication forks and also in repair of double-stranded DNA breaks by homologous recombination. It consists of the Smc5 and Smc6 proteins and the Nse1-6 protein subunits (Non-smc element). The Smc5 and Smc6 proteins interact via hinge domains on one side and contain ATPase head domains, which are able to dimerize in presence of 2 ATP molecules, on the other side. These head domains are also bridged by Nse4 subunit. Proper funtionality of the complex requires a topological binding to chromatin fiber, which is regulated by binding/hydrolysis of ATP and probably mediated by „opening“ of the complex via the interface between the Smc6/Nse4 and subsequent encircling of chromatin. In the bachelor thesis I focused on the Smc6/Nse4 and Smc5/Nse4 interaction interfaces. In this diploma thesis, I´m studying the interfaces between Smc6/Nse4 and Smc6/Nse3. Firstly, I expanded the mapped amino acid region on Smc6 protein. In the second part of the thesis, I investigated the viability and phenotype of S.pombe yeast with mutations of selected amino acids in Smc6 integrated into the yeast genome. Selected mutations interfere with the Nse4 or Nse3 interactions or affect the ATPase cycle between head domains in its various phases. In this work, we identified new amino acids on Smc6 protein surface responsible for interactions with Nse4 and Nse3 and characterized phenotypes of selected Smc6 mutations in the S.pombe genome.

Abstract:

Komplex SMC5/6 se účastní postreplikačních oprav DNA, stabilizace pozastavených replikačních vidliček a rovnež oprav dvouvláknových zlomů DNA homologní rekombinací. Skláda se z proteinů Smc5 a Smc6 a z proteinových podjednotek Nse1-6 (Non-smc element). Proteiny Smc5 a Smc6 na jedné straně interagují přes tzv. hinge domény a na opačném konci obsahují ATPasové hlavové domény, které mohou dimerizovat pomocí 2 molekul ATP. Tyto hlavové domény jsou také přemostěny podjednotkou Nse4. Pro správnou funkci komplexu je nutná topologická vazba na chromatinové vlákno, která je regulována vazbou/hydrolýzou ATP a zprostředkovaná pravděpodobně „otevíráním“ komplexu přes rozhraní mezi proteiny Smc6/Nse4 a následným obepnutím chromatinu. V rámci bakalářské práce jsem se věnoval charakterizaci interakčního rozhraní Smc6/Nse4 a Smc5/Nse4. V této diplomové práci se zabývám rozhraním mezi Smc6/Nse4 a Smc6/Nse3. Nejdříve jsem rozšíril mapovanou oblast aminokyselin proteinu Smc6. V druhé části jsem zjisťoval fenotyp a životaschopnost kmenů S.pombe kvasinek se začlenenými mutacemi vybraných aminokyselin Smc6 do jejich genomu. Boli vybrané mutace narušující interakci Smc6 s Nse4 nebo Nse3 a mutace zastavující ATPázový cyklus v jeho různých fázích. V této práci sa podařilo identifikovat další aminokyseliny proteinu Smc6 odpovídajíci za interakci s Nse4 a Nse3 a charakterizovali jsme fenotyp vybraných mutací Smc6 v genomu S.pombe.
