Analysis of chosen pathogenic microorganisms in small rodents

Abstract:

V této diplomové práci se věnujeme vybraným patogenním mikroorganismům, jejichž rezervoáry jsou malí hlodavci vyskytující se v České republice. Tyto vybrané patogeny způsobují zoonotická onemocnění, tedy onemocnění, jejichž agens jsou přenosná ze zvířat na člověka. Teoretická část diplomové práce v úvodu shrnuje obecné poznatky o šíření a vzniku infekčního zoonotického onemocnění u člověka včetně role hlodavců v tomto procesu. Dále se zde zabýváme několika onemocněními, k nimž se vztahuje praktická část práce, a to s cílem přiblížení kontextu jejich výskytu a možnostem diagnostiky u člověka. Jsou jimi lymeská borelióza, tularémie, Q-horečka a hantaviry způsobené infekce. Těmto zoonózám je zde věnována pozornost z hlediska infekčního agens, patogenity, průběhu infekce, diagnostiky, léčby, prevence a výskytu v ČR. Experimentální část práce má charakter studie výskytu (prevalence) patogenních mikroorganismů u odchycených hlodavců a je založena na detekci těchto infekčních agens v orgánech hlodavců nebo proti nim vytvořených protilátek v jejich séru a výplachu srdce. Metodou ELISA byla celkově detekována přítomnost specifických protilátek proti Borrelia burgdorferi s.l. v sérech v 7% hlodavců a ve výplaších ve 3,3%; proti Coxiella burnetii v sérech ve 9,5% a ve výplaších v 5,3%; proti Francisella tularensis v sérech ve 8,1% a ve výplaších ve 4,3%. Metodou nested RT-PCR byla detekována přítomnost hantavirové RNA v plicní tkáni hlodavců a z vyšetřených 98 vzorků byly hantaviry přítomny ve 3 případech.

Abstract:

In this thesis we study pathogenic microorganisms whose reservoirs are small rodents occurring in the Czech Republic. These selected pathogens cause zoonotic diseases whose agents are transmitted from animals to humans. The theoretical part of the introduction summarizes the findings about the spread and emergence of zoonotic infectious diseases in humans, including rodents role in this process. Furthermore, we are dealing with a number of diseases which are related to the practical part, with a view to approximate the context of their occurrence and diagnostic capabilities in humans. It includes Lyme disease, tularemia, Q-fever and infections caused by hantaviruses. These zoonoses attention is paid with regard to infectious agent, pathogenicity, during of infection, diagnosis, treatment, prevention and incidence in the country. The experimental part of the work has the character of a study of the incidence of pathogenic microorganisms in captured rodents and is based on the detection of these infectious agents in rodent organs or antibodies agains these patogens in their serum and heart rinses. ELISA was used to detect the presence of specific antibodies against Borrelia burgdorferi s.l. in sera of 7% in rodents and rinses in 3.3%; against Coxiella burnetii in sera in 9.5% and rinses in 5.3%; against Francisella tularensis against the sera of 8.1% and 4.3% in the rinses. Nested RT-PCR was used for detection of the presence of hantavirus RNA in rodent lung tissue. There were 3,1% (3 cases) occurence of hantaviruses in 98 examined samples.
