Expression and characterisation of AFL3 protein from Aspergillus fumigatus

Abstract:

V předchozích výzkumech bylo na našem pracovišti nalezeno 10 lektinům podobných proteinů Aspergillus fumigatus a po provedení bioinformatických studií byl vybrán předpokládaný lektin AFL3, který je kódovaný genem afl3. Tento gen byl již dříve na našem pracovišti naklonován do plasmidu a vložen do buněk Escherichia coli. Cílem bakalářské práce bylo pokusit se tento rekombinantní protein izolovat. Pracováno bylo tedy s buňkami Escherichia coli kmene BL21(DE3)/RIPL-pRSET-afl3 a BL21(DE3)/pLysS-pRSET-afl3. Prvním experimentem byl potvrzen fakt, že protein není exprimován v periplasmě a další experimenty se zabývaly izolací proteinu z média. Byly optimalizovány podmínky kultivace, nejlepších výsledků bylo dosaženo při pěstování kultury při 30°C přes noc. Na základě nízké detekovatelnosti našeho proteinu ve vzorku bylo zvoleno zakoncentrování médií, což se nejprve zdálo být vhodným postupem, při dalších experimentech bylo však dosaženo spíše neuspokojivých výsledků. Byly testovány různé typy médií, jako nejlepší se jevilo LB médium základního složení. Reverzibilní srážení proteinů z média síranem amonným bylo provedeno v celém spektru koncentrací síranu ve vzorku, většina předpokládaného proteinu byla vysrážena při 30 % nasycení. SDS-PAGE elektroforéza byla běžně prováděna na 15 % polyakrylamidovém gelu, následně byl použit koncentrovanější gel, který měl separovat proteiny o menších molekulových hmotnostech, uspokojivého výsledku však nebylo dosaženo. Jeden z možných problémů byla nízká kvalita stárnoucích buněk, z tohoto důvodu byla provedena izolace plasmidu pRSET-afl3 z buněk BL21(DE3)/RIPL-pRSET-afl3, tento plasmid byl transformován do čerstvých buněk Escherichia coli téhož kmene. Byly tedy připraveny nové buňky, se kterými mohou být prováděny další experimenty.

Abstract:

10 lectin-similar-proteins of Aspergillus fumigatus were previously discovered pursuant to a sequence homology. A formerly undescribed putative lectin AFL3 (encoded by the afl3 gene) was chosen. A DNA fragment containing afl3 was previously inserted into a cloning plasmid and a genetic transformation in the Escherichia coli was performed at our institute. The aim of this work was a possible reach up to a protein isolation. BL21(DE3)/RIPL-pRSET-afl3 and BL21(DE3)/pLysS-pRSET-afl3 were the two strains employed in the tests. The very first experiment – a simple way to prove the periplasmic localization of the protein - displayed the AFL3 absence in the periplasm. Consequently, a variety of media were used and optimized to provide a maximum quantity of the expressed protein. The optimum cultivation temperature should not exceed 30°C, the over-night growth was considered to be optimal. Due to the low protein production and only a little chance of detection, a media thickening was proposed to be a proper tool compromise. This procedure seemed to be an ideal way of getting closer to our goal. Hovewer, there had only been a little effect visible and it had not been stable. We optimized the medium type – usage of the LB medium was the best. The ammonium sulfate precipitation was realized in a broad range of ammonium sulfate concentrations. The AFL3 was best precipitated at a 30 % ammonium sulfate sample saturation. The commonly used SDS-PAGE polyacrylamide gel concentration was 15 %, protein samples were also separated using the 18 % polyacrylamide gel, although the results were not much satisfactory. One of the possible problems might had been a lower strain quality worked with, in that order, a pRSET-afl3 plasmid was isolated and transformed in new Escherichia coli cells. Accordingly, newly prepared Escherichia coli BL21(DE3)/RIPL-pRSET-afl3 is ready to be tested in the future experiments.
