Abstract:

Účinky koordinační sloučeniny, nanočástice a jejích syntetických konjugátů jsou velmi významné populární termíny v lékařské vědě. V dnešních mikrobiálních infekcích způsobených zejména bakterie odolné vůči více lékům jsou závažnými problémy pro lidskou společnost, v poslední době lékařskou vědou také čelí nejhoršímu scénáři u kmenů odolných vůči více lékům. Souřadnice sloučenin a nanočástice se široce používají k vývoji nových léčebných strategií proti bakteriální infekci. To již bylo řečeno, že Ruthenium koordinuje komplex a Ruthenium nanočástice, které mohou být použity jako baktericidní činidlo, avšak jejich účinky proti rezistentním patogenním bakteriálním kmenům nebyly plně studovány. V této studijní sérii konjugátu Ruthenium-Triazol Schiff báze bude syntetizován a aplikován na bakteriální kmeny rezistentní na antibiotika, aby se ověřily jeho biologické a farmakologické aktivity různými kroky optické hustoty, testy biologické aktivity, imunologické studie, mikroskopické techniky.Další důležitou pracovní částí je, zda syntetizované sloučeniny a QD mají protirakovinné vlastnosti nebo ne. Tato práce upozorňuje na účinky ruthenia v kombinaci se Schiffovou bází a triazolem na rezistentní bakteriální kmeny, rezistentní na vankomycin, rezistentní Staphylococcus aureus (VRSA) a meticilin rezistentní Staphylococcus aureus (MRSA). Na druhou stranu jsme se zaměřili na kvantové tečky a jejich roli v detekci a eliminaci bakterií. V této práci byly syntetizovány a použity proti různým kmenům bakterií dva různé typy teček Quantum. Syntetizované souřadnicové sloučeniny a QD studované v této práci mají vysokou účinnost proti těm rezistentním patogenním bakteriálním kmenům s nižším rizikem toxicity proti lidskému hostiteli. Pro charakterizaci syntetizovaných sloučenin a QD byly použity různé chemické charakterizační techniky jako FTIR, DLS, XPS, Raman atd.

Abstract:

Effects of the coordination compound, nanoparticle and its synthetic conjugates are very popular terms in medical science. In these days’ microbial infections especially caused by the multidrug-resistant bacteria are the serious problems to human society, recently medical science is also facing the worst scenario with multidrug-resistant strains. Coordinate compounds and nanoparticles are widely used to develop new treatment strategies against bacterial infection. It has been reported already that Ruthenium coordinate complex and Ruthenium nanoparticle can be used as a bactericidal agent however their effects against resistant pathogenic bacterial strains were not fully studied. In the present study series of Ruthenium-Triazole Schiff base conjugate will be synthesized and applied on antibiotic-resistant bacterial strains to check its biological and pharmacological activities by different optical density steps, biological activity assays, immunological studies, microscopic techniques. Another important work part is whether the synthesized compounds and QDs have anticancer property or not.The present thesis high lights the effects of Ruthenium combined with Schiff base and Triazole against resistant bacterial strains vancomycin-resistant Staphylococcus aureus (VRSA), and Methicillin-resistant Staphylococcus aureus (MRSA). On the other hand, we focused on quantum dots and their roles in bacterial detection and elimination. In this thesis, two different types of Quantum dots synthesized and used against resistant bacterial strains. The synthesized coordinate compounds and QDs studied in this thesis have high efficiency against those resistant pathogenic bacterial strains with lower toxicity risks against the human host. Different chemical characterization techniques like FTIR, DLS, XPS, Raman etc. were used for the characterization of the synthesized compounds and QDs.
