Biochemical markers of neurodegeneration

Abstract:

Tato bakalářská práce se zabývá biochemickými markery, které se vyšetřují při laboratorním vyšetření biologických vzorků pro zjištění přesnější diagnózy neurodegenerativního onemocnění. Popsány jsou zde také ty biomarkery, kterými se zabývají výzkumy a v praxi se ještě nepoužívají. Popis jednotlivých biomarkerů je zde rozdělen podle biologického materiálu, ze kterého jsou vyšetřovány - z mozkomíšního moku a z krve. V experimentální části práce byl zkoumán biomarker alfa-synuklein, který byl stanovován v mozkomíšním moku 65 pacientů s podezřením na neurodegenerativní onemocnění. K jeho analýze byla použita metoda EUROIMMUN Alpha-Synuclein ELISA. Měření proběhlo na automatickém analyzátoru EUROIMMUN Analyzer I ve FNO. Cílem jeho stanovení bylo zjistit, zda pomocí tohoto biomarkeru lze jednotlivé diagnózy neurodegenerativních nemocí od sebe rozlišit. Zaměření bylo především na Parkinsonovu nemoc a synukleinopatie (onemocnění spojené patologickým s ukládáním alfa-synuklenu v mozku). Dalším cílem bylo zjistit základní analytické charakteristiky použité metody ELISA. Naměřená data byla statisticky zpracována a nebyla potvrzena schopnost alfa-synukleinu rozlišit Parkinsonovu nemoc od zdravé populace. Získané výsledky si však odporují oproti dalším studiím. Dále byla zjištěna schopnost alfa-synukleinu rozlišit Alzheimerovu nemoc od zdravých jedinců. Tento výsledek je v souladu s jinými výzkumy.

Abstract:

This bachelor's thesis deals with biochemical markers that are investigated during the laboratory examination of biological samples to establish a more accurate diagnosis of neurodegenerative disease. Biomarkers that are being researched and are not yet used in practice are also described here. The description of individual biomarkers is divided here according to the biological material from which they are examined - from cerebrospinal fluid and from blood. In the experimental part of the work, the biomarker alpha-synuclein was investigated, which was determined in the cerebrospinal fluid of 65 patients with suspected neurodegenerative diseases. The EUROIMMUN Alpha-Synuclein ELISA method was used for its analysis. The measurement took place on the EUROIMMUN Analyzer I automatic analyzer in the FNO. The aim of its determination was to find out whether individual diagnoses of neurodegenerative diseases can be distinguished from each other using this biomarker. The focus was primarily on Parkinson's disease and synucleinopathies (diseases associated with pathological deposition of alpha-synuclein in the brain). Another goal was to find out the basic analytical characteristics of the ELISA method used. The measured data were statistically processed and the ability of alpha-synuclein to differentiate Parkinson's disease from the healthy population was not confirmed. However, the obtained results contradict other studies. Furthermore, the ability of alpha-synuclein to distinguish Alzheimer's disease from healthy individuals was found. This result is consistent with other research.
