Structural characterization of interaction between SOSS complex and RNA polymerase II

Abstract:

DNA dvouvláknové zlomy (DSB) jsou nejškodlivějším typem poškození DNA způsobujícím vysoce cytotoxické léze, které mohou ohrozit integritu genomu a podporovat tumorigenezi. Eukaryotické buňky vyvinuly opravný mechanismus nazývaný homologní rekombinace (HR), který opravuje poškozenou DNA a udržuje stabilitu genomu. Nedávné studie prokázaly, že transkripce ohrožuje integritu genomu tím, že vytváří podmínky pro akumulaci mutací a celkové chyby metabolismu DNA a má také jasný dopad na HR. V současné době je široce diskutováno chápání rolí RNA metabolismu a transkripce v opravách DSB. Jednou z potenciálních složek komunikace mezi opravou DNA a transkripcí je komplex SOSS. Ten obsahuje proteiny SSB, které se podílejí na detekci ssDNA, a jsou tak zásadní pro opravy poškození DNA a regulaci dalších opravných proteinů, v kombinaci s INTS, které vážou C-terminální doménu RNAPII a hrají roli v metabolismu RNA. V komplexech SOSS jsou tedy zastoupeny faktory podílející se na opravě poškození DNA, stejně jako faktory regulující transkripci. Další zkoumání těchto komplexů by tedy mohlo objasnit jeden z mechanismů, kterými jsou propojeny opravy DNA a transkripce. V této práci se zaměřujeme na přípravu proteinových komplexů SOSS1 a SOSS2. Podmínky exprese a purifikace byly optimalizovány. Proteinové komplexy byly purifikovány v dostatečné kvalitě a budou použity pro další strukturní charakterizaci jejich interakcí s RNAPII.

Abstract:

DNA double-strand breaks (DSBs) are the most deleterious types of DNA damage causing highly cytotoxic lesions that can endanger genome integrity and promote tumorigenesis. Eukaryotic cells have developed an error-free mechanism called homologous recombination (HR) that repairs the damaged DNA and maintains stability of the genome. Recent studies have proved that transcription compromise the genome integrity by creating conditions for accumulating the mutations and overall DNA metabolic errors, and has a clear impact on HR. However, the underlying molecular mechanism(s) remain(s) unclear. Nowadays, understanding the roles of RNA metabolism and transcription in processing DSBs repair is widely discussed. One of the potential components of cross-talk between DNA repair and transcription is SOSS complex. The SOSS complex contains SSB proteins participating in sensing of ssDNA which makes them crucial for DNA damage repair and regulating the focus formation of other supportive repair proteins, in combination with INTS proteins that bind C-terminal domain of RNAPII and play a role in RNA metabolism. The factors involved in DNA damage repair as well as those regulating transcription are represented in SOSS complexes. Therefore, further investigation of these complexes could clarify one of the mechanisms by which DNA repair and transcription are linked. In this thesis we focus on preparation of key reagents, protein complexes SOSS1 and SOSS2. The expression and purification conditions were optimised. Protein complexes were generated in sufficient quality and are used in further structural characterization of their interactions with RNAPII.
