Metabolic analysis of human pluripotent stem cells

Abstract:

Biologie kmenových buněk je základním kamenem regenerativní medicíny, neboť pacientům s degenerativními onemocněními skýtá naději na zlepšení či vyléčení jejich stavu. Recentní studie jasně ukazují, že způsob, jakým kmenové buňky využívají energii a nejrůznější intermediární metabolity, ovlivňuje jejich následný osud včetně schopnosti a směru diferenciace. Jedním z hlavních směrů současného výzkumu je proto identifikace klíčových hráčů metabolismu lidských pluripotentních kmenových buněk. Tato práce se proto zaměří na analýzu důležitých parametrů energetické homeostázy lidských pluripotentních kmenových buněk rostoucích v různých kultivačních podmínkách, což následně může napomoci definovat osud buňky během reprogramace a diferenciace. Zároveň bude také vytvořen počítačový model metabolismu kmenových buněk a to za využití experimentálních dat. Model bude následně experimentálně ověřen. Tato práce by tak měla přispět k pochopení regulace homeostázy lidských pluripotentních kmenových buněk, k vývoji nových strategií buněčné reprogramace, udržení pluripotence a efektivní diferenciace kmenových buněk.

Abstract:

Stem cell biology represents an essential cornerstone of regenerative medicine and provides the prospects of definitive solutions for patients with degenerative diseases, for which curative therapies are currently lacking. Nowadays emerging evidences indicate that the way in which stem cells use energy and intermediate metabolites determines their fate, including when they will differentiate and what they will become. The main aim of the current research is to identify the key metabolic players in human pluripotent stem cells. Accordingly, this study will focus on analysis of important parameters of energy homeostasis in human pluripotent stem cells under various culture conditions, which might define the cell fate during reprogramming and differentiation. Together with that, there will be established a computational model of stem cell metabolism using our experimental data for validation of the model. Consequently, this study will contribute to understanding the homeostasis in human pluripotent stem cells, and will add to development of new strategies to cellular reprogramming, maintenance of pluripotency and efficient differentiation of stem cells.
