Prevalence of Factor V Leiden in patients with positive ProC Global Test

Abstract:

Žilní trombózy jsou závažným zdravotním problémem.Leidenská mutace koagulačního faktoru V významně zvyšuje riziko trombózy.Zjišťovala jsem prevalenci této mutace v souboru pacientů sledovaných v hematologické ambulanci pro trombofilní stav.Důsledkem bodové mutace v genu pro faktor V je nahrazení argininu za glutamin v pozici 506 proteinu FV.Tato změna vede ke vzniku rezistence na aktivovaný protein C a ke zvýšené tvorbě trombinu.Vysoce citlivý screeningový test ProC Global jsem použila pro odhalení poruch v systému proteinu C.V případě pozitivity ProC Global byla vyšetřena mutace FV molekulárně biologickými metodami,dále byl vyšetřen protein C, protein S a další markery trombofílie.V souboru pacientů použitých v této práci jsem zjistila,že prevalence F V Leiden u pacientů s pozitivním ProC Global testem je 60,9%.

Abstract:

Venous thrombosis is a serious health problem.Coagulation factor V Leiden mutation significantly increases the risk of trombosis.I have examined the prevalence of this mutation in a group of patients followed in the haematology outpatient department for trombophilic state. The consequence of a point mutation in the gene for factor V is the replacement of glutamine with arginine at position 506 of protein F V.This change leads to an emergence of an activated protein C resistance and to an increased production of thrombin.A highly sensitive ProC Global screening test was used to defect faults in the protein C system.In case of ProC Global positivity FV mutation was examined by molecular biological methods. Protein C,protein S and other thrombophilia markers were examined as well. In the group of patients used for this study,the prevalence of FV Leiden in patients with positive ProC Global test was 60,9 %.
