Sensitivity of in vitro-cultivated plants to photoinhibition

Abstract:

Předkládaná diplomová práce se zaměřila na analýzu fotoochraných mechanizmů u rostlinných explantátů kultivovaných za různých intenzit záření a v různých kultivačních médiích pomocí indukované fluorescence chlorofylu. V práci analyzuji parametry fluorescence chlorofylu: FV/FM, ΦII, NPQ, qF0, qN, qI, qE a qT, které mohou být vypočteny z průběhu Kautského křivky indukované flouorescence chlorofylu a umožňují analyzovat jednotlivé primární fotosyntetické děje probíhající v pigment-proteinových komplexech v chloroplastu. V této diplomové práci byly jako modelové druhy použity druhy Potinara hybr. a Boswellia ameero. Cílem této práce byla kvantifikace fotochemických a nefotochemických procesů aktivovaných ve fotosyntetickém aparátu těchto rostlin v průběhu jejich ovlivnění dodatkovým zářením vyvolávajícím fotoinhibici. V práci jsou mezidruhově porovnány reakce na různá fotoinhibiční ovlivnění s cílem navrhnout vhodnou kultivační a experimentální hodnotu záření pro pěstování těchto rostlin in vitro.

Abstract:

The thesis focused an analysis of photoprotective mechanisms in plant explants cultivated under different light intensites on different cultivation media. The analysis exploited the technique of induced chlorophyll fluorescence, the following parameters in particular: Fv/Fm, ΦII, NPQ, qF0, qN, qI, qE, and qT. These chlorophyll fluorescence parameters can be calculated from Kautsky kinetics of chlorophyll fluorescence and help to evaluate individual primary processes of photosynthesis happening in chloroplastic pigment-protein complexes. In this diploma thesis, Potinara hybr. and Boswellia ameero were used as model plants. The aim of the thesis was to quantify photochemical and non-photochemical processes activated in plant photosynthetic apparatus when exposed to excess supplementary light doses causing photoinhibition. In the thesis, particular responses to sevral different photoinhibitory treatments are compared between specis so that suitable growth and experimental light doses might be recommended for each species when cultivated in vitro.
