Variability of gene phosphoethanolamine/phosphocholine phosphatase in chicken

Abstract:

Tato diplomová práce se zabývá variabilitou genu fosfoethanolamin/fosfocholin fosfatázy zapojeného do regulace mineralizace kostí u kura. Cílem práce bylo osekvenovat jednotlivé vzorky a vytvořit nukleotidové sekvence, ve kterých bylo ověřováno, zda je gen PHOSPHO1 variabilní ve sledovaném SNP ve třech populací kura. Teoretická část práce se zaměřuje na popis genu PHOSPHO1 a jeho vztahem k mineralizaci kostí. Dále se věnuje stručnému popisu kura, skeletu kura, genomu kura a genetickým polymorfismům, se kterými se úzce pojí molekulární markery. Pro tuto práci byly nejdůležitější SNP markery (jednonukleotidové polymorfismy), které byly vyhledávány pomocí molekulárně-genetických metod. Praktická část charakterizuje molekulárně-genetické metody, které dopomohly k dosažení výsledků práce. Tyto metody byly stručně popsány v teoretické části. Výsledky byly diskutovány a na jejich základě byl stanoven závěr.

Abstract:

This diploma thesis deals with the variability of the phosphoethanolamine/phosphocholine phosphatase gene involved in the regulation of bone mineralization in chickens. The aim of the study was to sequence individual samples and create nucleotide sequences in which it was verified whether the PHOSPHO1 gene is variable in the monitored SNP in three chicken populations. The theoretical part of the thesis focuses on the description of the PHOSPHO1 gene and its relation to bone mineralization. It also provides a brief description of the chicken, the chicken skeleton, the chicken genome, and the genetic polymorphisms with which molecular markers are closely related. The most important for this study were SNP markers (single nucleotide polymorphisms), which were searched using molecular genetic methods. The practical part characterizes the molecular-genetic methods that helped to achieve the results of the work. These methods were briefly described in the theoretical part. The results were discussed and based on them, a conclusion was reached.
