Působení platinového derivátu LA-12 na buněčnou linii HT-29 ve srovnání s cisplatinou a oxaliplatinou

The effect of platinum derivate LA-12 on cell line HT-29 in comparison to cisplatin and oxaliplatin

Abstract:

Cílem práce bylo studium účinku LA-12 ve srovnání s cisplatinou a oxaliplatinou na buňky kolorektálního karcinomu HT-29 v různých fázích buněčného růstu. Využili jsme proliferační křivky ke zjištění správné koncentrace buněk pro sledování rezistence závislé na konfluenci po aplikaci LA-12, cisplatiny a oxaliplatiny. Cytotoxicita byla detekována pomocí MTT testu mitochondriální aktivity podporovaným výsledky viability (barvení eosinem) a počtů buněk ve světelném mikroskopu (adherentní, plovoucí). K popisu buněčného cyklu jsme využili průtokovou cytometrii. Apoptózu jsme studovali detekcí procenta buněk s fosfatidyl serinem (annexin-V, flowcytometrie), snižení mitochondriálního potenciálu (TMRE, flowcytomwtrie), kondenzací a fragmentací jaderného chromatinu (DAPI, fluorescenční mikroskopie) a štěpení PARP (westerm blott). LA-12 vykazovala podobnou cytotoxicitu u obou studovaných populací (IC50 konfl. 4,52 µM, IC50 nekonfl. 4,42 µM). U oxaliplatiny byla zjištěna IC50 konfl 112, 56 µM a IC50 nekonfl. 6,9 µM, u cisplatiny nebyla IC50 konfl detekována vůbec a IC50 nekonfl. 18,8 µM. Při studiu buněčného cyklu vykazovaly všechny látky blok v S fázi s postupným přechodem do G2/M, jen u oxaliplatiny při koncentraci IC20 bylo zaznamenáno jen zpoždění ve fázi S s postupnou úpravou cyklu. LA-12 a cisplatina způsobovaly podobné navýšení počtu plovoucích buněk narozdíl od oxaliplatiny. Signifikantní změny v detekci fosfatidyl serinu a snížení membránového mitochondriálního potenciálu byly pozorovány u všech studovaných látek po 48 hod, ačkoli cisplatina vykazovala nejvyšší efekt. Ve stejném časovém intervalu měla LA-12 i cisplatina podobné výsledky při vyšetřování PARP a jaderné fragmentace ve srovnání s oxaliplatinou. LA-12 je schopna překonat rezistenci závislou na konfluenci u buněk HT-29 narozdíl od cisplatiny a oxaliplatiny. U rostoucích buněk způsobuje apoptózu. LA-12 vykazuje odlišný mechanismus aktivace apoptózy ve srovnání s oxaliplatinou i cisplatinou u buněk HT-29. Je u ní možné předpokládat pozdější využití v léčbě onkologických onemocnění.

Abstract:

The aim of this study was to investigace effects of LA-12 on colorectal carcinoma cell line HT-29 in comparison to cisplatin and oxaliplatin in other phases of cell growth. Subconfluent and growing population were used to study influence-dependent resistance after treatment with LA-12, cisplatin and oxaliplatin. Cytotoxicity was detected by MTT test of mitochondrial activity followed by viability (eosin) assay and detection of cell numbers (light microscopy- adherent and floating cells). Flowcytometry was used to describe changes in cell cycle. Apoptosis was evaluated as a percentage of cells with phosphatidyl serin (PS) externalization (annexin-V assay, flowcytometry), decrease of MMP (TMRE, flowcytometry), condensation and fragmentation of nuclear chromatine (DAPI staining, fluorescence microscopy) and PARP cleavage (western bloting). LA-12 had similar toxicity in both populations (IC50 confl. 4,52 µM or IC50 nonconfl. 4,42 µM respectively) in contrast to oxaliplatin (IC50 confl 112, 56 µM a IC50 nonconfl. 6,90 µM) and cisplatin (IC50 confl not detected or IC50 nonconfl. 18,80 µM). All compounds caused accumulation of cells in S phase and were further accompanied by increase in G2/M phase, only oxaliplatin (IC20) caused the transient delay in S phase with consequent repair of cell cycle. LA-12 and cisplatin caused significantly higher amount of floating cells than oxaliplatin. Significant changes in phosphatidyl serin externalization and decrease in MMP were detected after exposure to all compounds after 48 h. However, cisplatin had the strongest effect. At the same point, PARP cleavage and nuclear fragmentation were similar in cisplatin and LA-12 treated cells in contrast to oxaliplatin. LA-12is able to overcome confluent-dependent resistance of HT-29 cells to other platinum compounds. In growing cells it causes apoptosis but this cell death would be realized by different mechanism than that oxaliplatin and cisplatin. LA-12 is promising drug with supposed to use for treatment of cancer in the future.
