Mutation analysis of the promyelocytic leukemia gene

Abstract:

Gen PML kóduje multifunkční protein, který koordinuje důležité nádorové supresorické funkce, jako je zajištění stability genomu, opravy poškozené DNA, indukce apoptózy, zástava růstu a buněčná senescence. Potlačuje transkripční aktivitu promotorů specifických genů. Protein PML vytváří shluky, tzv. PML tělíska, v nichž interaguje s dalšími proteiny. Funkce proteinu PML je pravidelně narušena u akutní promyelocytární leukemie v důsledku chromozomální translokace t(15;17), jejímž důsledkem je narušení integrity PML tělísek. Inhibice funkce proteinu PML u akutní promyelocytární leukemie vede k nekontrolovatelnému růstu buněk a vzniku onemocnění. V souvislosti s akutní promyelocytární leukemií je gen PML poměrně dobře prostudován, avšak prozatím není známo onemocnění, které by bylo způsobeno zárodečnými mutacemi tohoto genu. Vzhledem k výše uvedeným funkcím proteinu PML by hypoteticky zarodečné mutace genu PML mohly predisponovat ke vzniku nádorových onemocnění. Cílem diplomové práce bude mutační analýza genu PML u pacientů s nádorovým onemocněním prsu nebo kolorekta s familiární zátěží. Analýza bude prováděna z genomické DNA metodou přímé sekvenace. V případě nálezu variant genu PML bude provedena populační studie za učelem zjištění frekvence těchto variant v normální populaci a rozhodnutí zda se jedná z klinického hlediska o polymorfismus či patogenní mutaci.

Abstract:

The PML tumour suppressor gene has become the object of the intense research owing to its involvement in the pathogenesis of acute promyelocytic leukemia (APL) where it is found fused to the retinoic acid receptor-a (RAR-a) gene as a consequence of the t(15;17) chromosomal translocation. The PML gene encodes PML protein, that is critical for the formation and stability of the PML-nuclear bodies (PML-NB). The PML protein and PML-NBs have been implicated in a number of cellular processes ranging from the cell growth and tumour suppression to apoptosis, transcriptional regulation, DNA-repair, cellular senescence and anti-viral defence. Reduced expression of the PML protein was described in solid tumours and this represents a selective advantage for growth of tumour cells. To date, we do not know any disease, that is caused by germline mutations of the PML gene. The aim of my work was to perform a mutation analysis of the PML gene in colon and breast cancer patients and to characterise the changes found in the PML gene sequence. I used direct sequencing. In the case, that I found the unknown variant, I made population study in the control group of non-cancer persons. I used the real time PCR method and high resolution melting (HRM) to detect the frequency of these novel variants in general population.
