Regulation of transcription and post-transcriptional modifications of mRNAs in plastids

Abstract:

The regulation of transcription is fundamental part underlying principles of central dogma of molecular biology. Yet, the mechanism of regulation of transcription in semi-autonomic plastids remains to be an unsolved puzzle. The synergy of the initial incorporation of plastids through primary endosymbiosis, the transfer of genes from plastids to the nucleus and subsequent serial endosymbiotic events resulted in distinct mechanisms of plastid gene expression and its regulation. The regulation of transcription of plastid genes can resemble that of their cyanobacterial ancestor or the eukaryotic (host) organism. The acquisition of plastids in organisms has led to the development of distinct traits at every level of transcription regulation, encompassing initiation, termination, and post-transcriptional modifications. As a result, there are significant variations in the mechanisms through which different plastid-bearing species control and regulate their gene expression. In this thesis, I compare, and contrast differences in regulation of transcription and post-transcriptional modifications in plastids and the prokaryotes. While classic model organisms like Arabidopsis or Chlamydomonas have been traditionally studied to understand molecular mechanisms of transcription regulation in plastids, euglenophytes such as Euglena gracilis and Euglena longa became valuable examples for investigating plastid transcription regulation due to their unique plastids and peculiar features in plastid metabolism. The remarks regarding newly acquired data are discussed in this thesis, with the focus on enabling future experimental studies that could illuminate the mystery behind transcription of regulation of plastids and unravel its molecular mechanisms.

Abstract:

Regulace transkripce je klíčovým prvkem základních principů centrálního dogmatu molekulární biologie. Přesto mechanismus regulace transkripce v semi-autonomních plastidech zůstává nevyřešenou záhadou. Synergie počáteční inkorporace plastidů prostřednictvím primární endosymbiózy, přenosu genů z plastidů do jádra a následných sériových endosymbiotických událostí vedla k odlišným mechanismům exprese genů plastidů a jejich regulaci. Regulace transkripce genů plastidů se může podobat regulaci jejich cyanobakteriálních předků nebo eukaryotickému (hostitelskému) organismu. Organismy nesoucí plastidy získaly také nové charakteristické vlastnosti na každé úrovni regulace transkripce, včetně iniciační fáze, terminace a posttranskripčních modifikací, což vedlo ke zvýšené diverzifikaci regulace transkripce genů plastidů na mezidruhové úrovni. V této práci porovnávám rozdíly v regulaci transkripce a posttranskripčních úpravách v plastidech a u prokaryot. Zatímco klasické modelové organismy jako Arabidopsis nebo Chlamydomonas byly tradičně studovány k porozumění molekulárním mechanismům regulace transkripce v plastidech, euglenofyti jako Euglena gracilis a Euglena longa se staly cennými příklady pro zkoumání regulace transkripce plastidů díky jejich jedinečným plastidům a zvláštnostem v plastidovém metabolismu. V této práci jsou diskutovány poznatky týkající se nově získaných dat se zaměřením na možné budoucí experimentální studie, které by mohly odhalit tajemství stojící za molekulárními mechanizmy regulace transkripce v plastidech.
