Determination of essential oil content by various methods in plant matrices

Abstract:

Diplomová práce se zaobírá různými metodami stanovení obsahu silic kmínu kořenného a srovnáním jejich výsledků. V literárním přehledu je představen kmín kořenný z hlediska pěstování, léčivých účinků a jeho využití v průmyslu. Jsou popsány silice, jejich účinky z obecného hlediska, ale také vztaženy ke kmínové silici. Dále jsou v práci představeny antioxidační, antibakteriální i antifungální účinky a složení kmínové nažky. V materiálu a metodice jsou pospány jednotlivé vzorky, lokality dodavatelských firem z ČR a metody stanovení vzorků. Množství silic bylo měřeno metodou destilace s vodní parou, superkritická fluidní extrakce a FT-NIR spektroskopie. Výsledky byly porovnány a statisticky vyhodnoceny. Mezi metodami nebyl potvrzen průkazný rozdíl v obsahu silic. Dále byla metodou dle Soxhleta extrakčně stanovena olejnatost kmínu. Výsledná olejnatost v sušině nažky byla 9,58-14,19 %. Pokusy byly provedeny u osmi vzorků odrůdy Aprim, Kamín, Prochan, Rekord, Lesix a neregistrovaného osiva merkantilu.

Abstract:

The diploma thesis deals with various methods of determining the content of essential oils of caraway seeds and comparing their results. In the literature review, caraway seeds are presented in terms of cultivation, medicinal effects and their use in industry. Essential oils are described, their effects in general, but also related to essential oil of caraway seeds. Furthermore, the work presents antioxidant, antibacterial and antifungal effects and the composition of caraway seeds. The material and methodology include description of particular samples, locations of supplier companies from the Czech Republic and methods for determining samples. The number of essential oils was measured using the method of steam distillation, supercritical fluid extraction and FT-NIR spectroscopy. The results were compared and statistically evaluated. No significant difference in the content of essential oil was confirmed between the methods. Furthermore, the oil content of caraway seeds was determined by extraction using the Soxhlet method. The resulting oil content in the dry matter was 9.58-14.19%. The experiments were performed on eight samples of the varieties Aprim, Kamín, Prochan, Rekord, Lesix and unregistered mercantile seeds.
