Abstract:

V posledních dekádách dochází k rychlému rozvoji velkého množství technik pro diagnostiku virů založených na nanotechnologiích, jako jsou nanočástice či kvantové tečky se širokou škálou povrchových modifikací. Disertační práce se zabývá návrhem a vývojem biosensorů pro detekci HPV (lidské papilomaviry) a HIV (viry lidské imunodeficience), které jsou vysoce nakažlivé a velmi často přenášené sexuálním kontaktem. V experimentální části práce byly pro rekognici a detekci těchto virů použity nanomateriály na bázi magnetických částic a kvantových teček. Tyto částice byly fyzikálně-chemicky charakterizovány a funkčnost biosensensorů byla validována pomocí PCR (polymerázové řetězové reakce) a elektrochemických analýz. Popsané biosensory poskytují levnou alternativu konvenčních methodik při zachování srovnatelné citlivosti a univerzálnosti, kdy je lze využít jako platformu pro detekci jiných virů či patogenů. Po detailní charakterizaci byly biosensory s úspěchem využity pro stratifikaci klinických vzorků pacientů s diagnostikovanými nádory hlavy a krku. Závěrem lze konstatovat, že biosensory mohou být efektivním diagnostickým nástrojem. Především díky možnostem modifikací a aplikovatelnosti v automatizovatelných systém by mohly být v budoucnosti uplatněny v klinické praxi.

Abstract:

In recent years, a large number of methods founded on identification and diagnostic of human viruses employing nanotechnologies containing, quantum dots, nanoparticles, and their surface modification have been consolidated. The present thesis describes specific biosensing system for viruses' detection, as HPV (human papillomavirus) or HIV (human immunodeficiency virus), being both high-risk viruses and the most commonly viruses sexually transmitted. The recognition and detection parts were defined and the biosensor was verified using magnetic isolation and fluorescence methodology and their confirmation was done through PCR (polymerase chain reaction) and electrochemical analysis. The developed sensor provides a cheap substitute to conventional technology with equal sensitivity and versatility characteristics than other methods and with a higher enhancement of nucleic acid detection for viruses or other pathogenic agents. Based on our results and after the characterization, we used our biosensor to stratify clinical specimens from patients who are suffering from head and neck cancer. It is right to conclude that our system can exert as an efficient instrument for diagnostics of the high-threat viruses. Because of their perfect properties and a possibility of easy improvements and execution into the automated trials, they could be used up for big scales in clinical level.
