Functional analysis of cells lacking Dishevelled
Abstract:
Tato diplomová práce ve své teoretické části shrnuje poznatky o Wnt signalizaci a proteinu Dishevelled, který je klíčovou komponentou této signalizace. V praktické části je využit buněčný model odvozený s buněčné linie HEK293, který byl vytvořen metodou CRISPR/Cas9 a chybí mu všechny isoformy proteinu DVL. Tento model by měl pomoci při studiu vlivu absence proteinu Dishvelled na Wnt signalizaci.
Abstract:
This thesis summarizes existing findings about Wnt signalling and protein Dishevelled that is a key component of these signalling pathways. In practical part, cell line model derived from HEK293 cells that was created by CRISPR/Cas9 and lacks all isoforms of DVL was used. This cell line should help to study influence of Dishevelled absence on Wnt signalling.
