Screening and identification ligninolytic enzymes of basidiomycetes fungi isolated from Czech forests

Abstract:

Ligninolytické enzymy se vyznačují schopností biodegradace a bioremediace. Jejich vlastnosti mohou být zkoumány v laboratorních podmínkách pomocí techniky kokultivace, která studuje vzájemné interakce organismů. Tyto interakce jsou důležitým faktorem pro produkci enzymů v kokultuře. Technika kokultivace byla použita v této práci v experimentálním skríningu, který je založený na posouzení růstového potenciálu kombinací ligninolytických hub.Do experimentu bylo zařazeno 19 kokultur bílé a hnědé hniloby. Pro detailní experiment byly vybrány tyto tři kombinace s nejlepšími výsledky: Laetiporus sulphureus/Trametes gibbosa, Piptoporus betulinus/Trametes gibbosa a Piptoporus betulinus/Stereum hirsutum.U nich byla spektrofotometricky měřena enzymová aktivita lakázy a lignin peroxidázy. Celková doba experimentu byla 35 dní, vprůběhu kokultivace bylo provedeno 5 paralelních odběrů v pravidelných intervalech 7 dnů.U enzymu lakázy byla největší produkce změřena ve 14. dnu kokultivace pro všechny testované vzorky. Kokultura Laetiporus sulphureus/Trametes gibbosa (LS/TG) vykazuje průměrně nejvyšší naměřené hodnoty aktivity enzymu lakázy. Nejvýraznější hodnota aktivity lignin peroxidázy byla pro všechny vzorky detekována až 21. den kultivace. Nejvyšší aktivitu lignin peroxidázy projevila také kokultura Laetiporus sulphureus/Trametes gibbosa (LS/TG). Enzymová aktivita lignin peroxidázy v celé délce měření dosahovala mnohem vyšších hodnot, než tomu bylo v případě aktivity lakázy.Zexperimentu vyplývá, že nejlepších výsledků pro oba enzymy dosahovala kombinace Laetiporus sulphureus/Trametes gibbosa (LS/TG). Proto byla aktivita lakázy i lignin peroxidázy této kokultury porovnána s aktivitou příslušných monokultur, tedy Laetiporus sulphureus (LS) a Trametes gibbosa (TG).Enzymová aktivita kokultury dosahovala v průběhu měření vyšších hodnot než aktivita monokultur. Kokulturu Laetiporus sulphureus/Trametes gibbosa (LS/TG) lze tedy považovat za slibnou pro další testování a případné praktické využití. Zvýšení aktivity enzymů u kokultury bylo s největší pravděpodobností dosaženo prostřednictvím interspecifických interakcí, které probíhají mezi kokultivovanými organismy.Výsledky této práce potvrzují závěry dosavadních vědeckých publikací, které říkají, že kokultivací vhodných vzorků lze zvýšit aktivitu enzymu. Nicméně pro využití v průmyslu je nutné tyto metody stále zdokonalovat a hledat další vhodné kombinace houbových kultur.

Abstract:

Ligninolytic enzymes are characterized by the ability of biodegradation and bioremediation. Their properties can be revealed under the laboratory conditions using the co-cultivation technique which studies the mutual interaction of organisms. These interactions are very important factor for the production of enzymes in the co-culture. The co-cultivation technique was used in this thesis as a part of experimental screening which is focused on the growth potential of ligninolytic fungi.Nineteen samples of white and brown rot fungi were tested during the experiment. Based on the experimental screening test three combinations of white and brown fungi (Laetiporus sulphureus/Trametes gibbosa, Piptoporus betulinus/Trametes gibbosa and Piptoporus betulinus/Stereum hirsutum) were selected for more detailed analysis.For these combinations, the enzyme activity of laccase and lignin peroxidase were determined spectrophotometrically. The total duration of the experiment was 35 days, during which 5 parallel samples were taken at regular intervals each seventh day.For all tested samples the highest activity of laccase was measured the 14th day of co-cultivation. On average, the highest values of laccase activity were measured for the co-culture Laetiporus sulphureus/Trametes gibbosa (LS/TG).Furthermore, the most significant activity of lignin peroxidase was detected on the 21st day of co-cultivation. The highest lignin peroxidase activity was detected also for co-culture Laetiporus sulphureus/Trametes gibbosa (LS/TG). The enzymatic activity of lignin peroxidase showed higher values in comparison with the laccase activity.In conclusion, the co-culture Laetiporus sulphureus/Trametes gibbosa (LS/TG), is very efficient for enhancing laccase and lignin peroxidase activity. Therefore, the activity of laccase and lignin peroxidase were compared with the respective monocultures - Laetiporus sulphureus (LS) and Trametes gibbosa (TG).The higher enzyme activity was detected for the co-culture compared with the monocultures. Therefore, the co-culture - Laetiporus sulphureus/Trametes gibbosa (LS/TG) could be attractive for further testing and eventual industrial application. The enhance enzyme activity was most likely achieved due to the mutual interactions in fungal co-culture.The results of this thesis confirm the conclusions of scientific publications which has been reported previously stating, that co-cultivation technique could be used for increasing enzyme activity. However, it is necessary to continue in improving co- cultivation technique and find another fungal combination for any future industrial use.
