Nitro-fatty acids as potential endogenous regulators of macrophage essential physiological functions.

Abstract:

Makrofágy, dlouho žijící myeloidní buňky imunitního systému, tvoří nedílnou součást tkání organismu. Z historického hlediska jsou makrofágy známy především díky své úloze v imunitní obraně. Nicméně se ukázalo, že vykazují velikou strukturní i funkční různorodost. I když jsou všechny fyziologické funkce makrofágů nezbytné pro rychlý vznik, rozvoj i ukončení zánětlivého procesu, většina z nich, dojde-li k jejich deregulaci, může vyvolat a zprostředkovat vážné patologické stavy, jako je ateroskleróza, plicní hypertenze, astma, fibróza či nádorové onemocnění. Z toho důvodu jsou dnes makrofágy považovány za vhodné terapeutické cíle. Zánětlivé a metabolické reakce jsou spojeny se zvýšenou hladinou reaktivních metabolitů kyslíku a dusíku. Vystavení endogenních nenasycených mastných kyselin (FAs) těmto molekulám vede, mimo jiné, k produkci nitrovaných mastných kyselin (NO2-FAs). Bylo ukázáno, že prostřednictvím vratné interakce s proteiny, např. receptory, transkripčními faktory a enzymy, vykazují NO2-FAs různorodé protizánětlivé i protioxidativní účinky. Díky tomu jsou NO2-FAs považovány za slibné terapeutické prostředky pro léčbu metabolických poruch a řady zánětlivých nemocí. Nicméně jejich přesný vliv na makrofágy není ještě zcela objasněn. Z toho důvodu bylo hlavním cílem této disertační práce zjistit účinek kyseliny nitro-olejové (NO2-OA) na diferenciaci makrofágů, jejich funkční specializaci a odhalit zapojené molekulární mechanismy. Objevili jsme, že fyziologicky relevantní koncentrace NO2-OA ovlivňují růstovými faktory indukovanou diferenciaci makrofágů a jejich proliferaci, což má za následek snížení počtu makrofágů i tvorbu jejich kolonií. Dále jsme ukázali, že NO2-OA významně zeslabuje funkční specializaci makrofágů charakterizovanou sníženou produkcí reaktivní molekul, zánětlivých cytokinů a expresí dalších znaků maturace a aktivace makrofágů. Předpokládáme, že zasahování NO2-OA do signálních drah spuštěných pomocí růstových faktorů a aktivátorů by mohlo představovat hlavní mechanismus, jakým NO2-OA ovlivňuje tyto procesy. Silný účinek NO2-OA byl zjištěn především na aktivaci proteinů transdukujících signály a aktivující transkripci, receptoru pro faktor stimulující kolonie makrofágů, mitogenem aktivovaných proteinových kináz, jaderného receptor kappa B a fosfatidylinositol-3-kinázy. Tato doktorská práce představuje NO2-FAs jako důležité signální molekuly zapojené do regulace různých fyziologických funkcí makrofágů. Dále přispívá k všeobecnému povědomí o rozličném působení NO2-FAs, které by mohlo být využito v léčbě chronických a zánětlivých onemocněních s komplexní patogenezí, včetně nemocí spojených s deregulovanými funkcemi makrofágů.

Abstract:

Macrophages are long-lived, myeloid immune cells that are strategically positioned throughout the body as essential components of mammalian tissues. They are historically known mainly for their role in host defense. Nevertheless, they exert great anatomical and functional diversity. Although all macrophage physiological functions are necessary for prompt development, progression, and resolution of inflammation, majority of them, when dysregulated, may provoke and mediate severe pathological states, such as atherosclerosis, pulmonary hypertension, asthma, fibrosis, and cancer. For this reason, macrophages are currently considered as attractive therapeutic targets. Inflammatory as well as metabolic reactions are associated with increased levels of reactive nitrogen and oxygen species. The exposure of endogenous unsaturated fatty acids (FAs) to these molecules results in generation of nitro-fatty acids (NO2-FAs), among others. NO2-FAs were shown to exhibit pleiotropic anti-inflammatory as well as anti-oxidative actions via reversible interactions with proteins, such as receptors, transcription factors, and enzymes. NO2-FAs are thus considered as a promising therapeutic tool for treatment of metabolic disorders and different inflammation-related diseases. Nevertheless, the exact mode of their action within macrophages remains to be fully elucidated. Therefore, the major aims of this Ph. D. thesis were to investigate the effect of nitro-oleic acid (NO2-OA) on macrophage differentiation, their functional specialization and to reveal molecular mechanisms behind NO2-OA actions. We discovered that physiologically relevant concentrations of NO2-OA affect growth factor-induced macrophage differentiation and proliferation, resulting in downregulation of macrophage numbers and colony formation. Additionally, we showed that NO2-OA significantly attenuates macrophage functional specialization, characterized by reduced production of reactive molecules, inflammatory cytokines, and expression of other markers of macrophage maturation and activation. We suggest that the interference of NO2-OA with growth factor- and activator-triggered signaling pathways could represent one of the major mechanisms how NO2-OA influences these processes. NO2-OA exerts strong impact particularly on activation of signal transducer and activator of transcription proteins, colony-stimulating factor receptor, mitogen-activated protein kinases, nuclear factor kappa B, and phosphatidylinositol 3-kinase. This Ph. D. thesis presents NO2-FAs as important signaling molecules involved in regulation of diverse macrophage physiological functions. Further, it contributes to general knowledge about the pleiotropic actions of NO2-FAs, which are considered suitable candidates for deployment against chronic and inflammatory diseases having complex pathogenesis, including those connected with dysregulated macrophage functions.
