Hmotnostně spektrometrické a fluorescenční zobrazování 3D buněčných kultur ošetřených perifosinem

Mass spectrometry and fluorescence imaging of perifosine-treated 3D cell cultures

Abstract:

Multimodální zobrazování je rozvíjející se obor medicinálně, biologicky a chemicky orientovaných věd. Vytváří velké množství informací, které jsou užitečné pro vysvětlení probíhajících procesů na molekulární, buněčné, tkáňové i tělesné úrovni. V mé práci bylo spojeno hmotnostně spektrometrické zobrazování pomocí matricí asistované laserové desorpce/ionizace s fluorescenční mikroskopií, a tyto dvě modality byly zkoregistrovány a matematicky vyhodnoceny. Jako biologický model rakoviny byly zvoleny 3D buněčné kultury neboli sferoidy, které napodobují mnohem přesněji chování nádorů, než klasické 2D buněčné linie. Pro jejich léčbu byl vybrán perifosin, což je sloučenina klinicky testovaná pro své protirakovinné účinky. Z analytického hlediska byl nejprve optimalizován protokol s fluorescenční analýzou sferoidů předcházející hmotnostně spektrometrickému zobrazování, ale tento postup byl omezen použitými fluorescenčními barvivy. Později byl úspěšně vyvinut protokol, který kombinuje hmotnostně spektrometrické zobrazování s následnou imunofluorescenční analýzou a zavádí algoritmy pro koregistraci a analýzu dat získaných ze stejného řezu sferoidu. Mnoho dalších biologických a biochemických měření perifosinu bylo provedeno biology pro zjištění jeho efektivity jako léčiva, ale nejsou zde popsány v detailu kvůli plánované publikaci.

Abstract:

Multimodal imaging is an emerging field of medically, biologically and chemically oriented sciences. It generates a great amount of extremely useful information for the explanation of ongoing processes at the molecular, cellular, tissue, or even body level. In my thesis, matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI MSI) was combined with fluorescence microscopy and the modalities were co-registered and mathematically evaluated. 3D cell cultures called spheroids, which mimic tumor behavior much more precisely than classic 2D cell cultures, were introduced as cancer models. For their treatment, a clinically tested drug with anticancer properties, perifosine, was selected. From the analytical point of view, first, the protocol for fluorescence imaging prior to MSI for multimodal spheroid analysis was developed; however, its further use was limited by the tested fluorescent dyes. Further, the protocol for subsequent MSI and immunofluorescence analysis of the same spheroid section was successfully developed, together with the image coregistration and data evaluation algorithm. Many biologist-provided biological and biochemical tests of perifosine effectivity were done but are not discussed here in detail as they are prepared for publication.
