Application of alkynes in organic synthesis

Abstract:

Oblast chemie alkynů, a zvláště pak alkynů terminálních, je v organické syntéze nezastupitelná a rozsáhlá. V teroretické části této práce je poskytnut pouze stručný výtah z chemie terminálních alkynů, které jsou studovány. Jsou shrnuty poznatky o bromoborační reakci acetylenu, esterifikace produktů bromoborace a příprava izomerních směsí stabilních cyklických boronátů. Je uvedena Negishiho a Sonogashirova cross-coupling reakce. Dále je poskytnut zevrubný přehled chránicích skupin pro vodík terminálních alkynů. Je představena kumarin-4-ylmethylová fotolabilních chránicí skupina (PPG). Popisuje se izolace (Z)-izomerů izomerních směsí stabilních cyklických boronátů a jejich využití v přípravě příslušných methyl, vinyl a fenyl derivátů pomocí Negishiho cross-coupling reakce. Další část této práce je věnována výběru molekuly vhodné pro Sonogashirovu cross-coupling reakci, v jejíž struktuře je přítomna terminální trojná vazba a fotolabilní chránicí skupina umožňující cílenou derivatizaci uhlíků této trojné vazby. Dále je popsána příprava nového esteru kyseliny propiolové chráněného fotolabilní (7-diethylaminokumarin-4-yl)methylovou odstupující skupinou. Jeho stabilita je diskutována ve vztahu k zamýšlenému užití v Sonogashirově cross-coupling reakci a cykloadiční reakci s organickými azidy.

Abstract:

Use of alkynes in organic synthesis is irreplaceable and utilization of terminal alkynes has unique standing within. This field is vast and diverse. Thus very short and focused excerpt of the field is given. The knowledge regarding acetylene bromoboration reaction, esterification of its products and preparation of isomeric mixtures stable cyclic boronates is compiled. Negishi and Sonogashira cross-coupling is introduced. Detailed survey of protective groups for the hydrogen of terminal alkynes is given. Photolabile coumarin-4-ylmethyl protective groups is presented. Isolation of (Z)-izomers from isomeric mixtures of stable cyclic boronates amd their utilization in preparation of respective methyl, vinyl and phenyl derivatives is described further. Next part of this work is dedicated to selection of suitable molecule for Sonogashira cross-coupling reaction containing terminal triple bond and fotolabile protecting group facilitating targeted derivatization of triple bond’s carbons. Preparation of propiolic acid ester protected with photolabile (7-diethylaminocoumarin-4-yl)methyl leaving group is accounted. Stability of the ester is discused in relation to intended use in Sonogashira cross-coupling reaction and in cycloaddition reactions with organic azides.
