Optimization of method for isoflavone and equol determination in cheese

Abstract:

Isoflavony, látky vyskytující se zejména v sóji luštinaté a jeteli lučním, a equol, hlavní metabolit isoflavonu daidzeinu, jsou v posledních letech zkoumány zejména pro jejich blahodárné účinky na lidské zdraví. A to nejen jako hormonální substituční terapie pro léčení příznaků spojených s menopauzou, ale také jako prevence proti rakovině prsu, kardiovaskulárním onemocněním, osteoporóze, úpravě hladiny cholesterolu a mnoho dalších pozitiv. Nejen jejich zdravotní účinky jsou popsány v teoretické části práce. Spolu s úvodním srovnáním fytohormonů a živočišných hormonů jsou v této části popsány i fenolické sloučeniny a fytoestrogeny, do kterých se isoflavony řadí. Samostatnou kapitolu poté tvoří equol, v níž je zahrnut také popis producentů a neproducentů equolu. Tuto část uzavírají kapitoly o výskytu těchto látek ve vybraných potravinách a teoretický popis úpravy vzorku pro isolaci isoflavonů a equolu ze sýra. Na tuto kapitolu navazuje experimentální část, kde je popsána pilotní metodika a výsledky pokusů stanovení vybraných isoflavonů a equolu dostupnou metodou, a to extrakcí do rozpouštědla a následnou analýzou pomocí LC-MS. V testovaném vzorku sýru byly nalezeny daidzein, glycitein, genistein a formononetin, oproti equolu a biochaninu A, které nalezeny nebyly.

Abstract:

Isoflavones which are especially contained in soya beans and red clover and equol, the main metabolite of isoflavone daidzein, have been studied for their positive impact on human health. They are the focus of interest thanks to their prevention function against many diseases such as breast cancer, high cholesterol, osteoporosis and treatment of symptoms associated with menopause. These effects and other topics included isoflavones and equol are described in the theoretical part of the thesis. There are also defined the phenolic compounds and phytoestrogens to which isoflavones belong and comparison between phytohormones and animal hormones. The separate chapter discus equol and the description of its producers and non-producers. The amount of isoflavones and equol in some food is mentioned at the end of that part. Also, the theoretical description of sample modification, which is followed by experimental part, where is the methodology and results of experiments are described to determine selected isoflavones and equivalents in a commercially available manner by extraction into a solvent and subsequent by LC-MS analysis. Daidzein, glycitein, genistein and formononetin were found in the tested cheese. Equol and biochanin A were not found in the sample.
