Study of influence of low-molecular metabolities on the expression of mRNA, which encodes enzymes of sarcosine metabolism

Abstract:

Má bakalářská práce pod názvem „Studium vlivu nízkomolekulárních metabolitů na expresi mRNA kódující enzymy metabolismu sarkosinu“ se zabývá studiem sarkosinu a jeho metabolismu spojeného se vznikem a rozvojem nádorového onemocnění prostaty. V dané práci byl studován vliv exogenního sarkosinu na expresi genů kódujících enzymy metabolismu sarkosinu, mezi které patří glycin N-methyl transferáza (GNMT), sarkosin dehydrogenáza (SARDH), sarkosin oxidáza (PIPOX) a dimethylglycin dehydrogenáza (DMGDH). Snažili jsme se zjistit, do jaké míry je ovlivněna exprese těchto genů u nenádorové prostatické linie PNT1A a dvou nádorových buněčných linií DU-145 a PC-3. Tato práce byla zahájena kultivací buněčných linií na specifickém médiu s přídavkem exogenního sarkosinu. Po 24. hodinové inkubaci byla provedena izolace RNA s následným ověřením její čistoty a integrity. Poté byla RNA přepsána do cDNA pomocí reverzní transkripce, a nakonec byla provedena relativní kvantifikace genové exprese pomocí qPCR s využitím interkalačního barviva SYBR Green. Výsledky byly statisticky zpracovány pomocí Studentova nepárového t-testu.Po aplikaci 10 µM sarkosinu jsme nepozorovali žádné signifikantní změny v expresi sledovaných genů. Až po aplikaci 50 µM sarkosinu jsme pozorovali signifikantní nárůst exprese SARDH, PIPOX a DMGDH, tedy genů, které se podílejí na degradaci sarkosinu. Tento efekt byl však zjištěn pouze u nenádorové linie PNT1A, nikoliv však u nádorové line PC-3.

Abstract:

My thesis is named „Study of influence of low-molecular metabolities on the expression of mRNA, which encodes enzymes of sarcosine metabolism.“ It deals with the study of sarcosine and its metabolism in connection with carcinogenesis of prostate. We studied an influence of exogenous sarcosine on the expression of genes, which encode enzymes of metabolism of sarcosine. These enzymes includes glycine N-methyl transferase (GNMT), sarcosine dehydrogenase (SARDH), sarcosine oxidase (PIPOX) and dimethylglycine dehydrogenase (DMGDH). We focused on detecting the influence of expression of these genes amongst several cell lines, particularly lines PNT1A, PC-3 and DU-145. Firstly we cultivated cell lines on a specific medium. After 24 - hours incubation, RNA isolation was performed. Then a quality of the isolated RNA was verified spectrofotometricaly. Also the integrity was checked by Bleach gel. Then, the RNA was transcribed into cDNA by reverse transcription. Then the relative quantification of gene expression was performed using qPCR reaction (with SYBR Green). In the end Student´s unpaired t-test was used for processing results.No significant changes in the gene expression were observed after application of 10 µM sarcosine. We observed a significant increase in the expression of SARDH, PIPOX and DMGDH after application of 50 µM sarcosine. This effects were observed only in the non-tumorous PNT1A line, but not in the PC-3 tumor line.
