Study of Metabolic Pathways of Biosynthesis of Biogenic Amines

Abstract:

Metody zabývající se detekcí a výzkumem vzniku biogenních aminů v potravinách nabývají v posledních letech na důležitosti, jak kvůli jejich negativním toxikologickým účinkům, tak kvůli jejich podílu na snížení kvality potravin. Tato práce se zabývá studiem metabolických drah biosyntézy biogenních aminů. V teoretické části je popsána charakterizace, význam i toxikologie aminů. Teoretická část obsahuje také přehledný popis metabolických drah tvorby polyaminů. Praktická část pojednává o návrhu vhodné sady PCR primerů určených pro detekci vybraných genů kódujících dekarboxylasy zodpovědné za produkci biogenního aminu putrescinu a rovněž optimalizaci této metody. Dále je zde prezentována optimalizace PCR metody pro použití nově navržených primerů a výsledky touchdown PCR. Správnost navržených primerů byla prokázána sekvenací.

Abstract:

Methods dealing with the detection and research of emergence of biogenic amines in foods become more important in recent years, both because of their negative toxicological effects, and because of their contribution to the reduction of the food quality. This work deals with the study of metabolic pathways in biosynthesis of biogenic amines. The theoretical part describes the characterization, importance and toxicology of amines. The theoretical part also provides a clear description of the metabolic pathways of polyamines formation. The practical part deals with the design of a suitable set of PCR primers designed for detection of selected genes coding decarboxylases responsible for the production of biogenic amine putrescine and also for optimizing PCR method for the usage of newly designed primers and touchdown PCR results. The accuracy of the proposed primers has been proved via sequencing.
