Use of chironomid larvae for evaluation of the aquatic environment pollution

Abstract:

Při relevantním posouzení ekologického stavu tekoucích vod nelze pominout toxicitu sedimentu. V odborné literatuře panuje stále živá diskuze jakými metodami a na jaké úrovni biologické komplexnosti toxicitu vyhodnocovat. Jedním za zajímavých jevů mohou být morfologické deformace zástupců makrozoobentosu, které jsou samy navrhovány jako možný nástroj biomonitoringu. Dosud byly intenzivně studovány u pakomárů. Přesto, že v terénu byly pozorovány časté korelace s koncentrací kovů a organochlorových pesticidů, četné snahy navodit deformace v laboratorních podmínkách skončily neúspěšně. V této diplomové práci byly vyhodnoceny frekvence deformací ústního ústrojí u larev pakomárů rodu Chironomus na osmi lokalitách s různým typem a mírou znečištění. Byly hledány korelace s ostatními markery toxického stresu (mortalita v kontaktním testu, intenzita detoxikačních procesů) a s koncentracemi vybraných polutantů. Kromě toho byla provedena série kontaktních laboratorních testů (akutní, subchronický a celoživotní) s modelovými látkami (kadmiem, fenantrenem a 2,4-dichlorfenolem). V těchto laboratorních testech byly hodnoceny stejné parametry jako v terénních studiích. Korelace prováděné na celém souboru žádný významný vztah mezi deformacemi a ostatními parametry nezaznamenaly. Srovnání lokalit na stejné řece nicméně hypotézu o souvislosti deformací a toxického stresu podporuje. V laboratorních experimentech se deformace navodit nepodařilo, s výjimkou případu, kdy byl design testu uzpůsoben pro vyšší realističnost přidáním krmiva před spikováním substrátu. Zdá se, že výsledky získané různými přístupy použitými v této studii neposkytují vždy shodné informace o studovaných lokalitách. Deformace ústního ústrojí mohou být v některých případech využity jako doplňkový nástroj bioindikace toxického znečištění.

Abstract:

In an ecological status assessment in running waters, the toxicity of sediment must not be omitted. Their is stil an intenzive discussion on the question of how to asses the toxicity and on which level of biological organisation this should be done. Morphological deformities in benthic animals are an interesting phenomenon, which is a potential biomonitoring tool as well. So far, they have been widely studied in mouthparts of Chironomid larvae. Despite the fact that there were many correlations between deformity frequencies and contamination described in the field, attempts to induce deformities in laboratory experiments were not successful. In this study, deformity frequencies in Chironomid larvae were analysed on 8 sites with different type and levels of contamination. Correlations with other markers of toxic stress (mortality in a contact test, intesity of detoxification) and with concentrations of selected polutants were examinated. In addition to that, a set of contact laboratory tests (an acute, a subchronical and a lifecycle tests) with selected contaminants (cadmium, phenanthrene, and 2,4-dichlorophenol) were performed. The same parametres as in the field studies were evaluated in these laboratory assays. Within the whole data set there were no signnificant correlations. Comparison of two sites on the same river, on the other hand, supports the assumption about a link between deformities and toxic stress. In the laboratory experiments there were no significant induction of deformities, with the exception of the case, when the test desing was changed in order to get more realistic conditions (the food was added before spiking the substrate). It seems that the results obtained in different approaches used in this study do not always give the same information about the studied locations. In some cases, mouthpart deformities might be used as a complementary tool for bioassessment of toxic contamination.
