Lectin Expression Possibilities in Eucaryotic System

Abstract:

Lektiny jsou proteiny neimunitního původu, které rozpoznávají a reverzibilně interagují se sacharidovými strukturami. Hrají důležitou roli v interakcích hostitelské buňky a patogenního organismu. Adhese patogenu na hostitelské tkáně je prvním impulsem k rozvoji bakteriální infekce. Tato práce se zabývá testováním genů čtyř předpokládáných lektinů Aspergillus fumigatus afl1, afl2, afl3 a afl3b. Byla provedena optimalizace PCR těchto genů. Každý gen byl amplifikován ve variantě se STOP kodonem a bez něj – pro připojení polyhistidinové kotvy, která může být využita pro afinitní purifikaci daného rekombinantního proteinu. Bylo provedeno jejich klonování genů do expresního vektoru pPICZα A, který slouží pro expresi rekombinantního proteinu v Pichia pastoris a pYES2/CT, využívaného pro expresi v Saccharomyces cerevisiae. pYES2/CT-afl1 byl elektroporací transformován do Saccharomyces cerevisiae. Následně byly provedeny expresní testy s transformovaným kmenem kvasinek S. cerevisiae INVSc1-pYES2/CT-afl1.

Abstract:

Lectins are proteins of non-immune origin, which recognize and reversibly interact with various saccharide structures. The play an important role in host-pathogen interactions. Adhesion of a pathogenic organism to a host tissue is a key impuls for the development of a bacterial infection. This thesis is focusing on testing of genes of four hypothetical lectins of Aspergillus fumigatus afl1, afl2, afl3 and afl3b. PCRs of these genes were optimized. Each gene was amplified in order to receive two constructs – with STOP codon and with the polyhistidine tag, which can be used for eventual affinity purification. The constructs were then cloned into pPICZα A expression vector, exploited for recombinant protein expression in Pichia pastoris and into pYES2/CT, used for expression and secretion of recombinant proteins in Saccharomyces cerevisiae. pYES2/CT-afl1 was electroporated into Saccharomyces cerevisiae. Subsequently, expression tests with transformed strain INVSc1-pYES2/CT-afl1 of Saccharomyces cerevisiae were performed.
