Thermal stability of nimesulide

Abstract:

Tato bakalářská práce se zabývá tepelnou stabilitou léčiva nimesulidu. Tepelná degradace nimesulidu byla studována v závislosti na různých rychlostech ohřevu pomocí kombinace termogravimetrie (TG) a diferenční termické analýzy (DTA). Při měření tepelného chování nimesulidu bylo použito devět rychlostí ohřevu. Hlavní cíl práce spočíval v popisu kinetiky tepelné degradace léčiva na základě TG křivek s důrazem na stanovení aktivační energie pro tento děj. Aktivační energie procesu degradace byla určena pomocí Kissingerovy metody a pomocí Ozawa-Flynn-Wallovy metody. Celková kinetika tepelné degradace byla popsána na základě Šesták-Berggrenova modelu, na jehož základě jsme vypočítali kinetické predikce. Následně byly kinetické predikce rozkladu léčiva nimesulidu diskutovány s ohledem na praktické zpracování tohoto léčiva v teplotním rozmezí 150-250 °C.

Abstract:

This Bachelor thesis deals with the thermal stability of the pharmaceutical nimesulide. Nimesulide was studied as a function of different heating rates using thermogravimetry (TG) in combination with differential thermal analysis (DTA). Nine heating rates were used to measure the thermal behaviour of nimesulide. The main goal of this work was to describe the kinetics of thermal degradation of the pharmaceutical based on TG curves with emphasis on the determination of the activation energy for this process. The activation energy of the degradation process was determined using the Kissinger method and the Ozawa-Flynn-Wall method. The overall kinetics of thermal degradation was described based on the Sestak-Berggren model, based on which kinetic predictions were calculated. Subsequently, the kinetic predictions for the degradation of nimesulide were discussed with respect to the practical processing of this drug in the temperature range of 150-250 °C.
