Anotace:

Problematika vztahu mezi onemocněním Lymeská borelióza, species Borrelia burgdorferi sensu lato, hlodavci (rezervoáry) a klíšťaty rodu Ixodes, je hlavním tématem předkládané diplomové práce. Její teoretická část se zabývá obecnými charakteristikami zoonóz, ke kterým Lymeská borelióza patří, ve vztahu k člověku a zvířeti. Dále poskytuje přehled různých vektorů, hostitelů a agens ve spojitosti se zoonózami. Následující část shrnuje nejnovější fakta a poznatky o Lymeské borelióze a její léčbě. Část experimentální dává přehled o použitých metodikách. Věnuje se polymerázové řetězové reakci jako nástroji amplifikace nukleových kyselin, která byla použita k zmnožení genu pro protein flagellin, aby mohla být určena pozitivita vzorků orgánů (ve výsledku pouze 0,59 % pozitivity, odpovídá jednomu vzorku). Hlavní část experimentu této práce zaujímá detekce specifických protilátek IgM a IgG proti Borrelia burgdorferi sensu lato pomocí nepřímé sendvičové metody ELISA. Na základě těchto zjištěných dat můžeme konstatovat, že výše pozitivity sér a výplachů získaných z vyšetřovaných jedinců odchycených hlodavců na B. burgdorferi sensu lato metodou ELISA, je pro IgM 10,17 % a pro IgG 0 %. Zprostředkovaně byla v této práci použita metoda mikroskopické aglutinace k detekci specifických protilátek proti konkrétnímu sérovaru Leptospira interrogans sensu lato ve vzorcích sér a výplachů. Zjištěna byla 5,08 % hraniční pozitivita sér hlodavců.

Abstract:

The problems of the relationship between the Lyme disease, the Borrelia species, rodents (the reservoirs) and the ticks of the Ixodes family, is the main topic of the presented diploma thesis. The theoretic part of the thesis is focused on the general characteristics of the zoonoses, among which the Lyme disease belongs, in relation to the humans and animals. Furthermore, the thesis provides an overview of different types of vectors, hosts and agents in relation to the zoonoses. The following part of the thesis summarizes the most recent facts and informations about the Lyme borreliosis and the possible ways of treatment. The experimental part of the thesis presents an overview of the methods applied. It deals with the polymerase chain reaction as a means of nucleic acid amplification, which has been used to multiply the gene for the flagellin protein in order to determine the positivity of the organ samples (the result has been positive in 0,59 %, a number that corresponds with one sample). The main body of the experimental part of the thesis lies in the detection of the specific IgM and IgG anti-bodies against Borrelia burgdorferi sensu lato by the means of the indirect Sandwich ELISA Method. Based on the available data, a conclusion was made – the percentage of the positive results of the sera and lavage, acquired from the individual caught rodents tested with the ELISA Method for Borrelia burgdorferi sensu lato, is 10,17 % for IgM, and 0 % for IgG. To detect the specific anti-bodies against a particular serovar Leptospira interrogans sensu lato in the samples of the sera and lavage, the method of microscopic agglutination has been used vicariously. A borderline positivity of 5,08 % has been detected in the rodent sera.
