Production of specific antibacterial agents by Salmonella enterica

Abstract:

Křížovým testem bylo testováno celkem 52 kmenů Salmonella enterica na produkci kmenově specifických antibakteriálních agens (tj. bakteriofágů, kolicinů, mikrocinů a sideroforů). Každý kmen byl tedy testován jako potenciální producent i indikátor. 14 kmenů (31 %) vykazovalo produkci bakteriofágů, 3 kmeny (6 %) produkovaly bakteriociny a 2 kmeny (4 %) byly schopny produkce jak bakteriocinů, tak bakteriofágů. Bakteriofágy salmonel byly přesněji charakterizovány podle morfologie plaků, spektra aktivity, inducibility mitomycinem C, restrikčního profilu fágové DNA a morfologie fágových částic získané elektronovou mikroskopií. Producenti bakteriocinů byli zkoumáni podle inhibičního spektra a morfologie inhibičních zón. Byla provedena PCR detekce 21 známých typů kolicinů a 7 mikrocinů, nebyl však prokázán žádný z nich. Je tedy možné, že tyto kmeny produkují nový typ antibakteriálního agens.

Abstract:

In a cross-test, we examined 52 strains of Salmonella enterica against each other as potencial producers of specific antibacterial agents (temperate phages, bacteriocins, microcins, siderophores) and as potencial sensitive indicators for them. 14 strains (31%) showed production of phages, 3 strains (6%) produced bacteriocins and 2 strains (4%) were capable of producing both agents – phages and bacteriocins. Salmonella phages were characterized by their morphology of plaques, host range, induction with mitomycin C, restriction endonuclease patterns and electron microscopy. Bacteriocin producers were analyzed by host range activity and morfology of the corresponding inhibition zones. None of the 21 tested types of colicins and 7 types of microcins yielded positive PCR reaction, suggesting that these strains could synthetize a new type of antibacterial agent.
