Chromosomal rearrangements with breakpoints in T cell receptor genes

Abstract:

T lymfocyty na svém povrchu exprimují αβ nebo γδ T-buněčný receptor (TCR) zodpovědný za jejich antigenní specifitu. TCR je kódován celkem čtyřmi geny - TRA, TRB, TRD a TRG, které se nachází ve třech různých chromozomových oblastech. Během maturace T lymfocytů jsou geny pro TCR přeskupovány místně-specifickou rekombinací mezi klastry variabilních (V), diverzifikačních (D) a spojovacích (J) segmentů. Tento proces označovaný jako V(D)J rekombinace predisponuje T lymfocyty ke vzniku aberantních přestaveb, především reciprokých translokací a inverzí. K nelegitimním rekombinacím mezi rozdílnými TCR geny dochází v malé míře také u zdravých jedinců a u lidí a myší je absolutní počet těchto přestaveb považován za ukazatel rizika vzniku lymfomu. Cílem této práce bylo určit frekvence nelegitimních rekombinací mezi TCR geny metodou fluorescenční in situ hybridizace s využitím malovacích sond a BAC konstruktů a jejich analýza sekvenováním PCR produktů u různých živočišných druhů včetně člověka. Výskyt chromozomových přestaveb s místy zlomů v TCR genech u prasat, skotu, ovcí, koní a čtyř dalších zástupců čeledi koňovitých byl zjišťován vůbec poprvé. Ukázalo se, že frekvence nelegitimních rekombinací mezi TCR geny se liší nejen při porovnání nálezů u různých živočišných druhů (a to i v případě vysoce příbuzných zástupců čeledi koňovitých), ale i v rámci druhu. Jsou zde diskutovány vlivy stáří vyšetřovaných jedinců, zastoupení γδ T lymfocytů v periferní krvi, komplexity, lokalizace a orientace TCR lokusů. Nemůžeme vyloučit ani souvztažnost mezi vysokým počtem aberantních přestaveb s místy zlomů v TCR genech u prasete, koně, skotu a ovcí a rizikem vzniku T buněčného lymfomu.

Abstract:

T cells express either αβ or γδ antigen-specific T cell receptors (TCR).There are four TCR genes - TRA, TRB, TRD and TRG located in three different chromosome regions. During maturation of T cells, TCR genes are rearranged by site-specific recombination between clusters of variable (V), diversity (D) and joining (J) segments. The process of V(D)J recombination predisposes T cells to aberrant rearrangements such as reciprocal translocations and inversions. Illegitimate recombination of different TCR genes occurs at a low level in healthy individuals and it has been suggested that the absolute number of these trans-rearrangement correlates with the risk of lymphoma in humans and mice. The objective of this work was to assess the frequencies of interlocus recombination between TCR genes by means of fluorescence in situ hybridization using painting probes and BAC probes and their analysis by sequencing of PCR products in different species including humans. The occurence of trans-rearrangements with breakpoints in TCR genes in pigs, cattle, sheep, horses and four other equids was investigated for the first time. We show that frequencies of illegitimate recombinations between TCR loci differ not only when comparing findings in different species (and even highly related species in case of equids) but also within the species. The effect of age of examined individuals, γδ T cells proportion in peripheral blood, complexity, localization and orientation of TCR loci are discussed here. The correlation between high number of trans-rearrangements with breakpoints in TCR genes in pig, horse, cattle and sheep and the risk of T cell lymphoma cannot be ruled out.
