The Influence of the Freezing Rate on the Enzymatic Activity

Abstract:

Cílem této práce byla studie vlivu rychlosti mražení a následného rozmražení na aktivitu enzymu ve vodném roztoku. Jako modelový enzym byl používán enzym laktát dehydrogenáza (LDH). Vliv rychlosti zmražení a následného rozmražení na obnovenou aktivitu LDH byl experimentálně studován bez přítomnosti kryoprotektantů. Enzym byl rychle zmražen v kapalném dusíku, pomaleji ve směsi suchého ledu s acetonem a při různých teplotách v kryostatu. Zjistilo se, že pomalé zmražení a rychlé rozmražení způsobuje menší ztrátu aktivity LDH, než rychlé zmražení v kapalném dusíku a pomalé rozmražení. Výsledky všech experimentů byly statisticky vyhodnoceny.

Abstract:

The aim of this work was to study the effect of the freezing and thawing rates on the activity of enzymes in aqueous solutions. Lactate dehydrogenase was used as model enzyme. The effect of freezing and thawing rate on the recovery of LDH activity was experimentally studied in the absence of cryoprotectants. Enzyme was frozen rapidly in liquid nitrogen, slowly in a mixture of dry ice with acetone and freezing at different temperatures in the cryostat. It was observed that slow freezing and rapid thawing caused less activity loss of LDH than rapid freezing in liquid nitrogen and slow thawing. The results of all the experiments were statistically evaluated.
