Identification and characterization of proteins regulated by plant hormones cytokinins by mass spectrometry

Abstract:

Rostlinné hormony cytokininy mají zásadní význam pro růst a vývoj rostlin. I přes intenzivní výzkum molekulární podstata působení cytokininů není dosud zcela objasněna. Zvláště pak o přímém vlivu cytokininů na změny proteinového profilu v rostlinách toho není dosud příliš známo. Pro proteomickou analýzu byli vybráni zástupci čtyř tříd cytokininů: hydroxylovaný isopentenylový, isopentenylový, aromatický typ a cytokininy odvozené od močoviny. Sedmidenní sazeničky huseníčku jarního (Arabidopsis thaliana) byly vystaveny krátkodobému působení cytokininů v kultivačním médiu. Po sběru rostlinek a následné extrakci proteinů, bylo využito metod gelové elektroforézy a hmotnostní spektrometrie s ionizací laserem za přítomnosti matrice v kombinaci s detektorem doby letu pro identifikaci proteinů, u kterých byla pozorována změna exprese vlivem cytokininů. Zkoumání a optimalizace různých proteomických metod vedly k podstatnému zlepšení identifikace a charakterizace rostlinných proteinů. Např. využití energie ultrazvuku a infračerveného záření umožnilo výrazné zkrácení času potřebného pro enzymatické štěpení proteinů prováděného před samotnou analýzou hmotnostním spektrometrem. Pro nalezení fosforylovaných proteinů a určení fosforylačních míst byly využity obohacovací techniky, jako např. afinitní chromatografie na imobilizovaných iontech a chromatografie s oxidem titaničitým. Sledování účinků exogenních cytokininů odhalilo 67 proteinů a 29 fosfoproteinů identifikovaných pomocí hmotnostní spektrometrie, u kterých bylo možné pozorovat výrazný vliv cytokininů na míru jejich exprese. Výsledky zde prezentované proteomické studie ukázaly mimo jiné možné propojení mezi vnímáním teplot rostlinou, signalizací pomocí vápenatých iontů a signálními drahami cytokininů.

Abstract:

Cytokinins are plant hormones that play an important role in plant growth and development. The molecular mechanisms of the cytokinin action are being intensely researched, but the changes of the proteome and phosphoproteome in response to cytokinin signals are still largely unknown. In order to get an insight into cytokinin signalling action at the protein level, 7-day-old Arabidopsis thaliana seedlings were treated with four main cytokinins representing hydroxyisopentenyl, isopentenyl, aromatic, and urea-derived type cytokinins. After total protein extraction with standard methods, gel electrophoresis followed by matrix-assisted laser desorption/ionization-time-of-flight mass spectrometry were used to separate and identify proteins that were differentially expressed under the cytokinin treatment. Various proteomic methods were examined to enhance the efficiency of the protein identification procedure. An ultrasonic energy and infrared radiation were applied to speed up and improve the enzymatic in-gel digestion of proteins. The use of enrichment techniques, such as immobilized metal ions affinity chromatography and titanium dioxide chromatography, was necessary to find the phosphorylation sites of phosphoproteins. After image analysis of the electrophoretic gels, reproducible significant differences between proteome and phosphoproteome patterns of Arabidopsis thaliana without and after cytokinin treatment were found for 53 and 31 spots, respectively. In these spots, 96 proteins were identified by mass spectrometry, revealing possible links between temperature, calcium, and cytokinin signalling.
