Histone characterization in pathogenesis of male infertility

Abstract:

During the process of spermatogenesis, male germ cells undergo a distinct morphological change and dramatic chromatin reorganization, whereby most histones are replaced by protamines, as part of the pathway to compact the genome into the small nuclear volume of the sperm head. Recent studies show the essential roles of epigenetic regulators, such as post-translational modifications (PTMs) and histone variants, in regulation of spermatogenesis and embryonic development following fertilization. Defects in either the replacement or the modification status of histones might cause male infertility. The main goal of the presented work was to introduce a methodology for histone characterization in sperm cells. Sample preparation based on microwave-assisted derivatization of histones using trimethylacetic anhydride in the presence of microwave radiation, followed by mass spectrometric analysis, made it possible to identify sequence variants and post-translationally modified forms of histones from patient samples with various anomalies in the spermiogram. Post-translational modifications of histones such as acetylation and methylation at lysine residues or phosphorylation at serine were quantitatively evaluated from the obtained proteomic data. The pilot proteomic analysis performed in the presented work is the basis for in-depth studies aimed at understanding the molecular mechanism that will allow the identification of epigenetic biomarkers of male infertility and may shed light on potential therapies for these patients in the future.

Abstract:

Během procesu spermatogeneze procházejí mužské zárodečné buňky zřetelnou morfologickou změnou a významnou reorganizací chromatinu, přičemž většina histonů je nahrazena protaminy za účelem zhutnění genomu v hlavičce spermie. Nedávné studie poukazují na zásadní roli epigenetických regulátorů jako jsou posttranslační modifikace histonů (PTM) a histonové varianty v procesu spermatogeneze a embryonálního vývoje po oplodnění. Defekty nahrazení nebo modifikačního stavu histonů mohou způsobit mužskou neplodnost. Hlavním cílem předkládané práce bylo zavézt metodiku pro charakterizaci histonů ze spermatických buněk. Příprava vzorku založená na chemické derivatizaci histonů anhydridem kyseliny trimetyloctové za účasti mikrovlnného záření, následovaná hmotnostně-spektrometrickou analýzou, umožnila identifikovat sekvenční varianty a posttranslačně modifikované formy histonů ze vzorků pacientů s různými anomáliemi ve spermiogramu. Ze získaných proteomických dat byly kvantitativně vyhodnoceny posttranslační modifikace histonů jako jsou acetylace a metylace na lysinových zbytcích, či fosforylace na serinu. Pilotní proteomická analýza provedená v předkládané práci je základem pro hlubší studie zaměřené na pochopení molekulárního mechanismu, které umožní identifikaci epigenetických biomarkerů mužské neplodnosti a mohou objasnit potenciální terapii pro tyto pacienty v budoucnosti.
