The differentiations of sugars content in plant material of higher plants under influence of enriched CO2 concentration and acclimated to different temperatures

Abstract:

Tato diplomová práce je zaměřena na aplikaci vytvořené metodiky využívající anthronového činidla pro simultánní stanovení nestrukturních sacharidů, škrobu a navíc také fotosyntetických pigmentů v experimentech studujících kombinovaný vliv oxidu uhličitého a ozářenosti, teploty a ozářenosti, a oxidu uhličitého a radiace na množství asimilátů v listech vyšších rostlin. V prvním experimentu bylo zjištěno, že zvýšená koncentrace oxidu uhličitého pravděpodobně vede k poklesu aktivity enzymu Rubisco spojené s nárůstem aktivity enzymu sacharosafosfátsyntasa (SPS), a proto k rychlejší syntéze sacharosy, která je především u mladších rostlin využita spíše pro transport do jiných sinků než pro jejich akumulaci v listech. Kromě nárůstu aktivity enzymu SPS dochází pravděpodobně také při zvýšení koncentrace oxidu uhličitého k nárůstu aktivity enzymů účastnících se degradace sacharosy (SuSy, syntasa sacharosy) a syntézy škrobu (ADP-glukosapyrofosforylasa), a proto k větší akumulaci škrobu v listech. Další experiment ukázal, že postupný vliv vysoké teploty a ozářenosti může stimulovat tvorbu asimilátů, ale při současném působení těchto dvou faktorů dochází k polední depresi fotosyntézy spojené s poklesem rychlosti fotosyntézy a tak poklesem rychlosti tvorby asimilátů. U rostlin aklimovaných na vysokou ozářenost a velmi vysokou teplotu (HI-HT2) byla polední deprese pozorována již v dopoledních hodinách, zatímco u rostlin vystavených vysoké ozářenosti a vysoké teplotě (HI-HT) polední deprese fotosyntézy pozorována nebyla. Naopak HI-HT rostliny vykazovaly ze všech variant rostlin největší množství asimilátů, a to ráno i dopoledne, spojované s tím, že v době odběru měly HI-HT rostliny optimální podmínky pozitivně stimulující aktivitu enzymů spojených s metabolismem sacharosy a škrobu, ale pravděpodobně i u těchto rostlin by s dalším nárůstem teploty a ozářenosti došlo k poklesu množství asimilátů způsobené polední depresí fotosyntézy. O kombinovaném vlivu zvýšené koncentrace oxidu uhličitého a difúzní složky radiace na obsah nestrukturních sacharidů a škrobu v listech rostlin toho není mnoho známo, proto lze pouze předpokládat, kombinovaný vliv těchto faktorů může pozitivně stimulovat aktivitu enzymů spojených účastnících se degradace sacharos (SuSy a invertasa) a syntézy škrobu (ADP-glukosapyrofosforylasa) a tudíž dochází v dopoledních hodinách k většímu nárůstu škrobu než nestrukturních sacharidů. Poslední část práce se zabývá studiem vlivu délky ohřevu na maxima absorpčních spekter a stabilitu zbarvení zelených (modro-zelených) kondenzačních produktů vznikajících reakcí sacharidů s anthronem v kyselém prostředí. Z testovaných hexos, ketohexosy poskytovaly velmi rychle maximální intenzitu zbarvení a při dalším zahřívání, docházelo k jejímu postupnému poklesu, aldohexosy reagovaly ze všech sacharidů nejpomaleji a poskytovaly nejstabilnější zbarvení. Pentosy, podobně jako ketohexosy, poskytovaly maximální intenzitu zbarvení velmi rychle, ale při dalším zahřívání docházeno k jejímu výraznému poklesu, který souvisí s nestabilitou zbarvení. Absorpční spektra všech testovaných hexos měla maxima absorpce při vlnové délce 625 nm, zatímco maxima testovaných pentos byla mírně posunuta ke kratším vlnovým délkám (604-606 nm).

Abstract:

This Diploma Thesis deals with the aplication of created anthrone method for the simultaneous determination of non-structural carbohydrates, starch and photosynthetic pigments, in addition, in few experiments studying combinated effect of carbon dioxide and irradiation, temperature and irradiation, and carbon dioxide and radiation on assimilates content in leaves of higher plants. In the first experiment, there was found out that elevated carbon dioxide concentration probably causes decrease activity of enzyme Rubisco associated with increase activity of enzyme sucrose-phosphate synthase (SPS, enzyme catalyzed sucrose synthesis) and thereby increase of sucrose synthesis rate which is especially in young plants rather used for the export to sinks than for the accumulation in leaves. Elevated carbon dioxide concentration probably also causes increase activity of enzymes associated with degadation of sucrose (SuSy, sucrose synthase) and synthesis of starch (ADP-glucose pyrophosphorylase) and thereby greater accumulation of starch in leaves. The next experiment showed that gradual high-irradiance and high-temperature effect can positive stimulate production of assimilates but combined influence of this two factors causes midday depression of photosynthesis associated with decrease of photosynthetic rate and decrease of assimilation rate. The midday depression of photosynthesis of HI-HT2 plants aclimated under high-irradiance and very high-temperature conditions was observed before midday whereas midday depression of photosynthesis of HI-HT plants aclimated under high-irradiance and high-temperature conditions was not observed. Of all types of plants, HI-HT plants contained the most of assimilates both in the morning and before midday due to optimal contitions during measuring which positive stimulated activity of enzymes associated with sucrose and starch metabolism. But futher increase of irradiation and temperature could cause midday depression of photosynthesis and decrease of assimilates content of this plants, too. There are not information about combined effect of elevated carbon dioxide and diffuse part of radiation on non-structural carbohydrates and starch content in leaves of plants and I can just suppose that combined effect of this two factors could positive stimulate activity of enzymes associated with degradation of sucrose (SuSy and invertase) and starch sythesis (ADP-glucose pyrophosphorylase) that is why there is greater starch content increase in comparison with non-structural carbohydrates content increase. The last part of this thesis deals with effect of different heating period on maxima of absorption spectra and stability of green (blue-green) condensation products which are created when carbohydrates are heated with anthrone in acid solution. Of the hexoses, ketohexoses gave maximum colour intensity quickly and marked gradual decrease in colour intensity with futher heating, aldohexoses reacted the most slowly with the most stable colour intensity, of all carbohydrates tested. Pentoses, as well as ketohexoses, gave maximum colour intensity quickly and marked pronounced decrease in colour intensity with futher heating which is associated with unstability of colour. Maxima of absorption spectra of all hexoses were at 625 nm whereas maxima of pentoses were shiffted towards shorter wave length (604-606 nm).
