Encapsulation of anticancer drugs in the cavity of ferritin nanocage

Abstract:

Nové možnosti terapie nádorových onemocnění jsou široce rozšířenou oblastí výzkumu. Do této oblasti léčby může být řazena i cílená terapie, která využívá nanočástic a jejich schopnosti navázání (tzv. enkapsulování) malých molekul do své dutiny a zamezení styku léčiva s necílovou tkání. Do kategorie proteinových nanočástic se zahrnuje i feritin, který v organismu za normálních okolností funguje jako přenašeč a zásobárna železa. Bohužel při procesu enkapsulace je možné, že se léčivo naváže i na vnější obal feritinu a nanočástice tak ztratí svoji účinnost jakožto bezpečný přenašeč léčiva nebo se při enkapsulaci naruší kvartérní struktura feritinu a léčivo z dutiny proteinu uniká. Proto byla tato práce věnována efektivní enkapsulaci za vysokých teplot bez narušení nativní struktury feritinu a odstraňování léčiva z povrchu proteinu pro získání ideálního nanopřenašeče k aplikaci v protinádorové terapii. Ze získaných dal lze vyvodit, že nejlepší výsledky vykazovala tepelná enkapsulace při 60 C s inkubačním časem 2 h, kdy se enkapsulovalo průměrně u tří šarží 478,5 molekul Dox s velikostí nanopřenašeče 11,7–18,2 nm a u šarže PfuFn(MCH145), kde proběhlo odmytí Dox nejslibněji u vzorků se zeta-potenciálem −28,4 mV (15,7 nm) a −28,6 mV (11,7 nm) v prostředí s 0,001M EDTA a 0,05M EDTA.

Abstract:

The treatment of cancer is a widespread area of research. It is possible to include targeted therapy into this area, which employs nanoparticles and their ability to bide (or encapsulate) small molecules into their cavity and separate the drug from contact with off-target issue. Ferritin, belonging to the group of protein-based nanoparticles, is normally present as a carrier and reservoir of iron in the body. Unfortunately, during the encapsulation process, it is possible that the drug attaches also to the outer shell of the ferritin and the nanoparticles lose their relevance as a safe drug carrier, or the encapsulation breaks down the quaternary structure of the ferritin and the drug leaks from the protein cavity. Therefore, this work was devoted to effective drug encapsulation at high temperatures without disturbing the native structure of ferritin and removal of the drug from the surface of the protein to obtain an ideal nanocarrier for application in anticancer therapy. From the obtained data it can be concluded that the best results were shown by thermal encapsulation at 60 C with an incubation time of 2 h, when encapsulated on average in three batches 478.5 Dox molecules with a nanocarrier size of 11.7–18.2 nm and in batch PfuFn (MCH145), where Dox was most prominently washed away for samples with zeta-potential −28.4 mV (15.7 nm) and −28.6 mV (15.7 nm) in 0.001M EDTA and 0.05M EDTA environment.
