Genome integrity control in prostate cancer stem cells

Abstract:

prostaty je nejčastěji diagnostikovaným typem rakoviny u mužů a jednou z nejčastějších příčin nádorově specifických úmrtí v rozvinutých zemích. Teoretická část této diplomové práce je zaměřena na teorii kancerogeneze, mechanismy DNA oprav, charakteristiku karcinomu prostaty, možné formy jeho léčby s důrazem na inhibitory PARP a histonových deacetyláz a rovněž na výzkum v oblasti kmenových buněk rakoviny prostaty. Experimentální část práce se zabývá ucelenou charakteristikou buněčné linie DU-145 a také testováním několika dalších modelových linií karcinomu prostaty. Z výzkumu vyplývá, že buněčná linie DU-145 byla ve srovnání s PC-3 odolnější vůči kombinaci ozáření a PARP inhibicí. Inhibitor histonových deacetyláz SAHA způsobil snížení exprese mutovaného p53 u linie DU-145 a rovněž lepší odpověď na radioterapii. Vůči ozáření byla více odolná CD24 pozitivní subpopulace DU-145. Výše uvedené poznatky přispívají k řešení problematiky personalizované protinádorové terapie, ale zároveň vyžadují další výzkumnou práci v této oblasti.

Abstract:

Prostate cancer is the most frequently diagnosed cancer in men and one of the most common causes of cancer-releated deaths in developed countries. The theoretical part of the thesis is focused on the theory of carcinogenesis, DNA repair mechanisms, characteristics of prostate cancer, possible forms of therapy with an emphasis on PARP inhibitors and histone deacetylases as well as on the current knowledge on prostate cancer stem cells. The experimental part is focused on a comprehensive characterization of cancer cell line DU-145, and also on testing of several other cell lines derived from prostate cancer. The results suggested that the cell line DU-145 is more resistant to a combination of irradiation and PARP inhibition compared to cancer cell line PC-3. Histone deacetylase inhibitor SAHA decreased expression of mutated p53 in DU-145 which led to better response to radiotherapy. The CD24 positive subpopulation of DU-145 was more resistant to radiation exposure than negative subpopulation. These findings are relevant to the tailoring of personalized cancer therapy, however, they also require further research.
