The possibility of eliminating the negative effect of hatching egg storage on hatchability

Abstract:

Tato bakalářská práce se zabývá vlivem preinkubace na líhnivost dlouhodobě skladovaných vajec. Pro pokus č. 1 bylo použito 9600 ks násadových vajec hybridní kombinace Dekalb White, jež byla skladována 23 (kontrolní skupina) a 21 dnů (pokusná skupina). Polovina vajec byla 7. a 14. den skladovacího období zahřívána na cílovou teplotu 95 °F a 60 min při této teplotě ponechána. U 60 vajec bylo stanoveno také stádium vývoje embryí. Pro pokus č. 2 bylo použito 8700 ks násadových vajec rodičovského hejna Moravia BSL, která byla rozdělena do čtyř skupin; první skupina byla skladována 22-25 dnů (P1), druhá skupina 17-21 dnů (P2), třetí skupina 14 dnů (P3) a čtvrtá 9-12 dnů (P4). Vejce skupiny P1 a P2 byla 11. a 18. den skladování preinkubována stejně jako u pokusu č. 1.U pokusu č. 1 byl vyřazený počet vajec při prosvěcování statisticky průkazně (P˂0,05) nižší u pokusné ve srovnání s kontrolní skupinou. U pokusné skupiny byla také statisticky průkazně (P˂0,05) nižší embryonální mortalita a vyšší oplozenost. Ve srovnání s nezahřátými vejci nezpůsobilo první ani druhé zahřátí průkazné zvýšení stádia vývoje embryí.U pokusu č. 2 byl počet vyřazených vajec nejnižší ve skupině P4. Statisticky průkazně (P˂0,05) vyšší počet vyřazených vajec byl pouze ve skupině P3, u které byl i nejvyšší podíl neoplozených vajec, jež byl v porovnání s P1 a P4 statisticky průkazně (P˂0,05) rozdílný. Časná embryonální mortalita byla průkazně (P˂0,05) nejvyšší u P1. Pro snížení časné embryonální mortality u dlouhodobě skladovaných vajec (14 dnů) byla délka preinkubace 3 až 3,5 hodin nad teplotu 28 °C dostačující. Posun ve vývoji embryí u hybridní kombinace Dekalb White není pro zlepšení líhnivosti při dlouhodobém skladování žádoucí. Opakovaná preinkubace dlouhodobě skladovaných vajec eliminovala negativní dopad skladovaní při líhnutí jak finálních hybridů, tak i plemenného materiálu.

Abstract:

This bachelor thesis deals with the influence of preincubation on hatchability of long-term stored eggs. For Experiment 1, 9600 hatching eggs of the Dekalb White hybrid combination were used and stored for 23 (control group) and 21 days (experimental group). Half of the eggs were heated to the target temperature of 95 ° F in 7th and 14th day of the storage period and left at this temperature for 60 minutes. In 60 eggs also the stage of embryonic development was determined. For Experiment 2, 8700 hatching eggs of the parent flock of Moravia BSL were used. These eggs were divided into 4 groups; the 1st group was stored for 22-25 days (P1), the 2nd group for 17-21 days (P2), the 3rd group for 14 days (P3) and the 4th for 9-12 days (P4). The 11th and 18th day of storage eggs of group P1 and P2 were preincubated as in Experiment 1.In Experiment 1, the number of eggs discarded at candling was statistically significantly (P˂0.05) lower in experimental group in comparison with control group. There was also statistically significant (P˂0.05) lower embryonic mortality and higher fertilization in experimental group. Compared to unheated eggs, neither the first nor the second heating caused a significant increase in the stage of embryonic development. In Experiment 2, the number of discarded eggs was the lowest in group P4. The statistically significant (P˂0.05) higher number of discarded eggs was only in the group P3, in which the highest proportion of unfertilized eggs was, which was statistically significantly (P˂0.05) different compared to P1 and P4. Early embryonic mortality was significantly (P˂0.05) highest in P1. To reduce early embryonic mortality in long-term stored eggs (14 days), preincubation period of 3 to 3.5 hours above 28 ° C was sufficient.To improve hatchability during long-term hatching egg storage a shift in embryonic development for the Dekalb White hybrid combination is not desirable. Repeated preincubation of long-term stored eggs eliminated the negative impact of storage during hatching process of final hybrids, as well as breeding material.
