Contribution to the theory and practice of capillary zone electrophoresis

Abstract:

Předkládaná disertační práce je zaměřená na teoretické a experimentální studium elektromigračních jevů indukovaných vzorkem nebo lokální změnou pH během elektroforetické analýzy. Tyto jevy ovlivňují separační účinnost, citlivost, správnost a reprodukovatelnost metody. Disertace je založená na souboru 8 prací, z nichž sedm je již publikováno v recenzovaném impaktovaném časopise Electrophoresis, osmá je do tisku přijata. Byla popsána existence a vlastnosti nového typu migrujícího rozhraní, tzv. hybridního rozhraní, které může vznikat i v jednoduchých základních elektrolytech často používaných v CZE praxi. Toto rozhraní složené z ostré a disperzní části ovlivňuje vývoj tvaru zóny vzorku a tím tvar detegovaného píku. Byly zkoumány principy zakoncentrování (stackingu) analytů pomocí zóny komponenty vzorku mající hybridní rozhraní a dán teoretický popis a matematická kritéria pro kvantitativní stacking. Byly popsány vlastnosti elektroforetických pohybujících se rozhraní v systémech obsahujících vícesytné slabé elektrolyty, kde neplatí Kohlrauschův regulační princip. Vznikají zde zóny nečekaných vlastností a koncentrační adjustace nemůže být předpovězena na základě dosud používaných znalostí. Dále byly popsány existence a vlastnosti komplexních hybridních systémových rozhraní, která mohou existovat v jednoduchých elektroforetických základních elektrolytech s vícesytnou slabou kyselinou, např. ve fosfátovém pufru. Byly definovány základní typy a principy stackování. Podrobně byla zkoumána jedna z příčin situací, kdy je detegováno více píků než je počet komponent vzorku. Tyto píky zůstávají často nevysvětleny či označeny za nečistoty či systémové píky. Jedním ze zdrojů těchto efektů jsou vzorky s vysokým obsahem solí (např. biologické vzorky), kdy makrokomponenta ze vzorku způsobí rozštěpení či deformaci zóny mikrosložky analytu v důsledku přechodných efektů či lokálních změn pH. Objev inverzního elektromigračně-dispersního profilu umožnil vypracovat metodu pro fokusaci neamfoterních slabých ionogeních analytů. Tento princip je odlišný od stackování i od IEF. Byl popsán vliv vzdušného oxidu uhličitého absorbujícího se do základního elektrolytu o vysokém pH během analýz s nepřímou UV absorbanční a vodivostní detekcí, kde vzniklé karbonáty indukují řadu nežádoucích systémových efektů a přechodných stavů, které způsobují silné změny v migraci analytů a jejich detekci. Všechny teoretické závěry a předpovědi získané z počítačových simulací a rychlostních diagramů byly ověřeny experimentálně.

Abstract:

This PhD thesis is aimed at theoretical and experimental study of electromigration effects induced by the sample or local pH changes during CZE analyses. These effects influence the separation efficiency, sensitivity and method accuracy and reproducibility. The thesis is based on 8 papers, 7 of them has been published in the impacted journal Electrophoresis, the last one has been submitted. The existence and properties of a new type of migrating zone boundary, the so-called hybrid boundary, were described. This type of boundary (composed of self-sharpening part and electromigration dispersion part) which may be formed even in simple BGEs and therefore be quite frequent in CZE practice, may affect significantly the evolution of shapes of sample zones, and consequently the shapes of detected peaks. The principles of stacking of analytes by a zone of a bulk sample component that possesses one hybrid boundary were investigated. A theoretical description of such a system was given and general rules and mathematical criteria for quantitative stacking were presented. The properties of electrophoretic moving boundaries in systems containing multivalent weak electrolytes where the Kohlrausch regulating law is not valid were studied. It was shown that zones of unexpected properties can be formed in such systems and that the adjusted concentrations in these zones cannot be predicted using present knowledge. The existence and properties of complex hybrid system boundaries were described and it was shown that this type of boundaries can exist even in simple electrophoretic BGEs with multiprotic weak acids, e.g., in phosphate buffer. Basis types and principles of stacking were defined in a comprehensive way. One of the reasons was investigated, where more peaks than the number of sample components are detected. Such peaks are often left unexplained or assigned to unidentified impurities or system peaks. It was shown that one source of such effects are samples with high salt content (e.g. biological samples), where a sample macrocomponent evokes splitting or deformation of the analyte zone as a result of transient effects or local pH changes. The influence of carbon dioxide from air absorbed in highly alkaline BGEs during analyses with indirect UV absorption and conductivity detection was studied. The forming carbonate induces a number of unwanted system effects and transient states, which induces strong and pronounced temporary changes in the migration of analytes and their detection. All theoretic results and predictions obtained from computer simulations and theoretical calculations were verified by experiments. The discovery of inverse electromigration dispersion profiles made it possible to develop a method for focusing of nonamphoteric weak ionogenic analytes. This principle is different from both CZE stacking and IEF.
