Study of reaction of adamantane-1-carbonyl chloride with benzylmagnesium chloride

Abstract:

Mezi deriváty isochromanonu patří celá řada přírodních i syntetických látek s významnými biologickými účinky, avšak je popsáno pouze malé množství obecných re-akcí přípravy těchto derivátů. Jednou z reakcí kde isochroman-1-on vzniká je titulní reakce adamantan-1-karbonylchloridu s příslušnými benzylmagnesium halogenidy. V návaznosti na předchozí výzkumné aktivity spojené s bakalářskou prácí, byl zjišťován vliv reakčních podmínek ? počáteční koncentrace výchozích látek, délka reakční doby a přítomnost dal-ších látek v reakční směsi ? na průběh reakce adamantan-1-karbonylchloridu s 3-methylbenzylmagnesiumchloridem. Všechny zmiňované podmínky ovlivňují ať už pozi-tivně nebo negativně, průběh této reakce, ale jednoznačný důkaz o mechanismu vzniku isochromanonových derivátů se nepodařilo získat. Proto byla provedena izotopová analýza pomocí značeného [18O]adamantan-1-karbonylchloridu s vyhodnocením distribuce značení v produktech reakce. Pomocí ESI-MS analýzy byla navržena fragmentace značeného 3-(1-adamantyl)-6-methyl-3-(3methylbenzyl)[18O]isochroman-1-onu a po provedení mode-lových reakcí byl následně navrženo několik alternativ mechanismu vzniku isochromano-nových derivátů. Mimo isochromanonových derivátů byla studována struktura dalších pro-duktů reakce 1,3-dionů, pomocí instrumentálních metod (RTG, IR, NMR a Ramanovy spektroskopie).

Abstract:

Many natural and synthetic substances with significant biological effects belong to the family of the isochromanone derivatives. Nevertheless, a small number of general reac-tions leading to these substances is described. As a continuation of previous work, the in-fluence of reaction conditions ? initial concentrations of starting compounds, reaction times and some additional components in reaction mixture ? upon the composition of crude products of the reaction of 1-adamantane carbonyl chloride with 3-methylbenzylmagnesium chloride were studied. All these conditions affect either positively or negatively the course of examined reaction, but definite evidence of the mechanism of isochroman-1-derivatives was not received. Therefore an isotope analysis was carried out by using [18O]adamantane-1-carbonyl chloride with the monitoring of labelling distribution in reaction products. Incorporation of one oxygen from starting [18O]adamantane-1-carbonyl chloride was observed and the position of labelled oxygen was investigated using ESI-MS tandem mass spectra analysis. Having carried out model reactions, several possi-ble mechanistic pathways leading to the isochromanone were suggested. Using the instru-mental methods (NMR, IR, MS, X-ray), the structure of 1,2,3-trisubstituted-1,3-diones ? other interesting molecules arising in the studied reaction ? was investigated.
