Preparation of labeled lectins for bioanalytical purposes

Abstract:

V této diplomové práci se věnuji lektinům bakterie Burkholderia cenocepacia, a to BC2L-A a BC2L-C-ct. Cílem této práce bylo připravit značené lektiny, sledovat vlastnosti značených a neznačených lektinů, a následně fluorescenčně značené lektiny využít. Značené lektiny se běžně využívají ke studiu glykosylací bakteriálních povrchů. Oba tyto lektiny byly purifikovány a poté úspěšně naznačeny FITCem. Byly porovnávány vlastnosti značených a neznačených lektinů pomocí SPR během dialýzy a poté pomocí ITC. S lektiny bylo provedeno několik mikroskopických experimentů, kde byla pozorována jejich vazba na povrchy s mannosou nebo heptosou. Fluorescenčně značené lektiny byly úspěšně využity na nově vyvíjené „glycan array.“ Dále jsem se v této práci pokusila vypurifikovat BCL-C-nt, což je další lektin B. cenocepacia, který je markerem pluripotentních buněk.

Abstract:

In this diploma thesis, I focus on lectins of bacterium Burkholderia cenocepacia, namely BC2L and BC2L-A-C-ct. The aim of this work was to prepare labeled lectins, observe the properties of labeled and non-labeled lectins, and then use the fluorescently labeled lectins. Labeled lectins are commonly used to study the glycosylation of bacterial surfaces. Both lectins were purified and then successfully labeled by FITC. The properties of labeled and non-labeled lectins were compared using SPR during dialysis and then using ITC. Several microscopic expreriments have been done with the lectins, where their binding was observed on surfaces with mannose or heptose. Fluorescently labeled lectins have been successfully used on "glycan array." Further, I tried purify BC2L-C-nt, another lectin of B. cenocepacia, which is a marker of pluripotent cells.
