Identification of tumor-specific T cells by T cell receptor beta analysis

Abstract:

Diplomová práce je zaměřena na možnosti identifikace nádorově-specifických T lymfocytů. V teoretické části popisuje dostupné metodiky identifikace, v praktické části se pak zaměřuje na konkrétní metodu založenou na studiu T lymfocytárního receptoru beta – na metodu TRBV PCR. Cílem práce je zavedení této metody do laboratorní praxe, její standardizace, testování na reálných vzorcích a porovnání jejích výsledků s výsledky získanými již používanou metodou anchor PCR.

Abstract:

This thesis is focused on possibilities of tumor-specific T cells identification. It describes known identification methods in theoretical part as well as one particular method of identification by T cell receptor beta analysis – TRBV PCR method – in practical part. The aims of this thesis are introduction TRBV PCR into laboratory field, standardization of this method, testing on real samples and comparison between outcomes of this method and outcomes of the other method – anchor PCR.
