The role of L-arginine, methylarginines and their metabolism in the endothelial homeostasis regulation

Abstract:

Cílem této diplomové práce bylo popsání vlivu argininu a methylargininů na udržování vaskulární endoteliální homeostasy. Pomocí metody Western blot byl sledován vliv hypoxie (1% O2) a asymetrického dimethylargininu na expresi a fosforylaci endoteliální syntázy oxidu dusnatého a protein kinázy B, dále byl sledován vliv na expresi transkripčního nukleárního faktoru κB, inhibitoru transkripčního nukleárního faktoru κB, hypoxií indukovatelného faktoru 1 a intercelulární adhezní molekuly 1 u in vitro pěstovaných lidských plicních arteriálních endoteliálních buněk. Dále byla u těchto buněk pomocí metody ELISA stanovena exprese endotelinu 1, míra proliferace pomocí ATP-celulárního testu a produkce oxidu dusnatého pomocí Griessovy kolorimetrické metody. Byl také sledován vliv argininu, asymetrického dimethylargininu, symetrického methylargininu a monomethylargininu na in vitro pěstované lidské umbilikální žilní endoteliální buňky, kde byla následně pomocí metody Western blot sledována exprese a fosforylace endoteliální syntázy oxidu dusnatého a protein kinázy B, dále byl sledován vliv na expresi transkripčního nukleárního faktoru κB, inhibitoru transkripčního nukleárního faktoru κB a intercelulární adhezní molekuly 1.

Abstract:

The aim of this thesis was to describe the effect of arginine and methylarginine in maintaining vascular endothelial homeostasis. With the help of Western Blot the effect of hypoxia (1% O2) and asymmetric dimethylarginine on expression and phosphorylation of endothelial nitric oxide synthase and protein kinase B, also, the effect on the expression of nuclear factor-κB transcription, a transcriptional inhibitor of nuclear factor κB, hypoxia-inducible factor-1  and intercellular adhesion molecule-1 with in vitro cultured human pulmonary artery endothelial cells were observed. Further, there was the expression of endothelin -1 determined in these cells, with the help of ELISA method, the rate of proliferation with the help of ATP cellular test and nitric oxide production using Griess colorimetric method. Also the effect of arginin, asymetric dimethylarginin, symetric methylarginin and monomethylarginin was watched with in vintro cultured on human umbilical vein of endothelial cells, where subsequently the expression and phosphorylation of endothelial nitric oxide synthase and protein kinase B were studied using Western blot, furthemore, the effect on the expression of the transcription nuclear factor κB inhibitor of transcription nuclear factor κB and intercellular adhesion molecule 1 were watched.
